摘  要:目的探讨β-葡聚糖对巨噬细胞源性泡沫细胞胆固醇逆转运的影响及相关机制。方法采用氧化低密度脂蛋白(ox LDL)诱导的RAW264.7巨噬细胞为泡沫细胞模型,给予β-葡聚糖进行干预。通过油红O染色及酶比色法定量检测细胞内总胆固醇和胆固醇酯水平的变化;采用Western blotting及实时定量PCR检测β-葡聚糖对巨噬细胞ATP结合盒转运蛋白A1(ABCA1)及其m RNA表达的影响并进行统计分析。同时采用蛋白合成抑制剂放线菌酮(CHX)处理细胞4、8、12 h测定ABCA1稳定性。结果与ox LDL处理组比较,β-葡聚糖对巨噬细胞摄取脂质的抑制率为(90.0±1.8)%,ox LDL处理组巨噬细胞源性泡沫细胞内胆固醇酯及总胆固醇与β-葡聚糖处理组(2.5、5.0、10.0μg/m Lβ-葡聚糖+ox LDL共同孵育)比较,差异均有统计学意义(P<0.05);5.0、10.0μg/m Lβ-葡聚糖处理组巨噬细胞源性泡沫细胞ABCA1及其m RNA表达与对照组比较,差异均有统计学意义(P<0.01或0.05)。β-葡聚糖(10.0μg/m L)与CHX共同处理组在细胞孵育4、8、12 h时,ABCA1降低水平均小于CHX处理组,差异有统计学意义(P<0.05)。结论β-葡聚糖通过增加ABCA1的表达水平及增强其稳定性,促进细胞内胆固醇的排出,最终抑制巨噬细胞源性泡沫细胞的形成。Objective To explore the effects of β-glucan on the reverse cholesterol transport of macrophage-derived foam cells and its relative mechanism. Methods Oxidized low-density lipoprotein(ox LDL) induced RAW264.7 macrophages were adopted as the foam cells model for giving the β-glucan intervention. The Oil-red O staining and enzymatic colorimetry were employed to examine the changes of intracellular total cholesterol and cholesteryl ester;Western blotting and real-time PCR were used to detect the effect of β-glucan on the expression on ATP-binding cassette transporter protein A1(ABCA1) and m RNA in macrophages. The obtained data were statistically analyzed. The protein synthesis inhibitors cycloheximide(CHX) processed cell4,8,12 h,thus,deter minated ABCA1 stability. Results The inhibiting rate of β-glucan on macrophage′ lipid uptake was(90.0±1.8)%,total cholesterol and cholesteryl ester levels had statistical difference between the ox LDL group and the co-in cu bation groups of ox LDL and 5.0,10.0 μg/m L β-glucan(P〈0.05);the expression of ABCA1 and m RNA had statistical differences between the 5.0,10.0 μg/m L β-glucan groups and the control group(P〈0.01 or 0.05). β-glucan(10.0 μg/m L) and CHX common treatment group in cell incubation 4,8,12 h,ABCA1 lower level less than CHX treatment group,the difference was statistically significant(P〈0.05). Conclusion β-glucan promotes the intracellular cholesterol discharge and finally inhibits the formation of macrophage-derived foam cells by enhancing ABCA1 expression and protein stability.
