摘 要:目的利用COI序列对市售鹿茸饮片、鹿茸粉进行鉴定,并建立统一的分子鉴定方法。方法对不同产地的鹿茸饮片及鹿茸粉分别进行DNA提取、COI序列扩增和序列测定,建立分子鉴定方法,并对市售鹿茸饮片、鹿茸粉进行调查分析。结果鹿茸饮片均可以使用COI通用引物进行PCR扩增和测序;物种之间相互区分明显;市售40份药材中14份为药典规定物种,26份为非药典规定物种;鹿茸粉未成功获得COI序列。结论COI序列的DNA条形码鉴定方法可准确、有效鉴定市售鹿茸饮片,此方法为市场监管鹿茸饮片提供科学手段。Objective Using the COI sequences of commercially available identification pilose antler,deer antler powder slices,and establish the unified molecular identification method. Methods The different origin of pilose antler yinpian and pilose antler powder respectively for DNA extraction,amplification and sequencing COI sequences,molecular identification method,and the commercial antler yinpian,velvet antler powder were investigated. Results All can be used antler yinpian COI universal primers for PCR amplification and sequencing; Distinguish between species is obvious; In the sold 40 that 14 pieces are the pharmacopoeia in medicinal species,26 species are the pharmacopoeia regulations. Velvet antler powder successful COI sequences. Conclusion COI sequences of DNA barcode identification method is accurate and effective identification of pilose antler yinpian,this method for market supervision,provide a scientific means of velvet antler slices.
