摘 要:目的研究芦丁对糖尿病小鼠的降血糖作用。方法采用四氧嘧啶(45 mg·kg-1)诱导糖尿病小鼠模型。通过观察糖尿病小鼠一般症状,检测糖尿病小鼠糖耐量,血清中空腹血糖、空腹胰岛素、胰岛素敏感指数,白介素-6、白介素-1β和肿瘤坏死因子-α的表达以及肝脏丙二醛、过氧化氢酶、谷胱甘肽过氧化物酶和超氧化物歧化酶的活性,评估芦丁对糖尿病小鼠的降血糖作用。结果芦丁呈剂量依赖性改善糖尿病小鼠全身症状和糖耐量,增高胰岛素敏感指数和空腹胰岛素水平以及过氧化氢酶、谷胱甘肽过氧化物酶和超氧化物歧化酶活性,降低血清中空腹血糖、白介素-6、白介素-1β和肿瘤坏死因子-α水平以及肝脏丙二醛含量。结论芦丁可通过抗氧化应激和炎性反应而减轻胰岛素抵抗,进而减轻糖尿病。Objective To investigate the hypoglycemic effect of rutin in diabetic mice. Methods A model of diabetes in mice was induced by alloxan (45 mg.kg- 1 ). The general symptoms, sugar tolerance, levels of fasting blood glucose ( FBG), fasting insulin ( FINS ), insulin sensitivity indexes ( ISI ), interleukin-6 ( IL-6 ), interleukin-1 β ( IL-1 β ) and tumor necrosis factor- α ( TNF- α ) in serum, the activity of malondialdehyde ( MDA ), catalase (CAT) ,glutathione peroxidase(GPx) and superoide dismutase(SOD) in the liver of diabetic mice were exanimed to evaluate the hypoglycemic effect of rutin in diabetic mice. Results Rutin improved general symptoms, sugar toler- ance and increased the levels of FINS, ISI and the activities of CAT, GPx and SOD in diabetic mice in a dose-de- pendent manner,reduced the levels of FBG, IL-6, IL-1β and TNF- α and the content of MDA. Conclusion Rutin can decrease the level of blood glucose in diabetic mice, which is associated with improved insulin resistance by suppressing oxidative stress and inflammation.
