加味七方胃痛颗粒对胃癌癌前病变大鼠CDK2/4的干预效应

摘　　要：目的:探讨加味七方胃痛颗粒对胃癌癌前病变(precancerous lesions of gastric cancer,PLGC)大鼠CDK2/4基因的干预效应。方法:采用化学致癌物N-甲基-N'-硝基-N-亚硝基胍(N-methyl-N-nitro-N-nitroso-guanidine,MNNG)建立胃癌癌前病变动物模型,采用免疫组化检测CDK2/4基因表达,Western blot法检测各组大鼠PLGC胃黏膜组织CDK2/4的表达水平。结果:与模型组比较,加味七方胃痛颗粒组和胃复春组的胃黏膜萎缩程度、胃黏膜肠化生程度明显减轻;加味七方胃痛颗粒组、胃复春组CDK2/4基因蛋白表达显著降低(P<0.01);相对于胃复春组,加味七方胃痛颗粒CDK2/4蛋白表达显著降低(P<0.01)。结论:加味七方胃痛颗粒可能是通过下调CDK2/4基因蛋白的表达,改善胃黏膜病理变化,抑制细胞分裂与增殖,从而干预PLGC进展。Objective： To observe the effect of Modified Qifang Weitong Granule on CDK2 /4 genes in treating for rat＇s precancerous lesions of gastric cancer. Methods： animal model of precancerous lesions was first established by using N-methyl-N-nitro-N-nitrosoguanidine（ MNNG）. CDK2 /4 gene expressions were detected with immunohistochemistry method. The expression levels of CDK2 /4 in rats＇ PLGC gastric tissue were detected with Western blot method. Results： Compared with those of the model group,the degrees of gastric atrophy and gastric intestinal metaplasia significantly relieved and the CDK2 /4 gene and protein expression was significantly lower（ P 〈0. 01） of Qifang Weitong Granule group and Weifuchun group. Compared with that of the Weifuchun group,the CDK2 /4 protein expression of Qifang Weitong Granule group was significantly lower（ P 〈0. 01）. Conclusion： Modified Qifang Weitong Granule can achieve better clinical efficacy by reducing CDK2 /4 gene and protein expressions,improving the conditions of gastric mucosa,inhibiting cell division and proliferation,and eventually preventing the progress PLGC.
