摘 要:目的:观察肾系康全方及其益气活血组、滋阴清热组2个拆方组对系膜增生性肾小球肾炎(mesangial proliferative glomerulonephritis,Ms PGN)大鼠肾组织α平滑肌肌动蛋白(Alpha smooth muscle actin,α-SMA)、肝细胞生长因子(hepatocyte growth factor,HGF)的影响,以明确该方及2个组成部分发挥的作用强弱,并探讨其部分作用机理。方法:以雄性SD大鼠为研究对象,采用隔日口服牛血清白蛋白并尾静脉注射葡萄球菌肠毒素B及皮下分点注射完全氟氏佐剂和不完全氟氏佐剂的方法复制Ms PGN模型,应用免疫组化法检测各组大鼠肾组织中α-SMA、HGF表达水平并进行半定量分析。结果:肾系康方与2个拆方组均可降低肾组织中α-SMA表达,上调肾组织中HGF表达,与模型对照组比较,差异有统计学意义(P<0.05);拆方组中,益气活血组明显优于滋阴清热组,两组比较,差异有统计学意义(P<0.05);各治疗组作用强度为全方组>益气活血组>滋阴清热组。结论:肾系康方能够抑制肾组织α-SMA的表达,上调HGF的表达,可能是其防治肾小球硬化和肾脏纤维化的作用机制之一;肾系康方及其拆方均有明显作用,益气活血组明显优于滋阴清热组,而全方组更为显著。Objective: To observe the effect of Shenxikang on mesangial proliferative glomerulonephritis( Ms PGN),Alpha smooth muscle actin( α-SMA) and hepatocyte growth factor( HGF) and to illustrate the mechanism of the process. Methods: In this study,male SD rats were used as the research objects. Every other day with oral use of bovine serum albumin and intravenous injection of staphylococcal enterotoxin B and by complete subcutaneous points injection and incomplete subcutaneous point injection Ms PGN model rats were made. Immunohistochemical method was used to detect the renal tissue α-SMA,HGF expression of rats and semiquantitative analysis was made. Results: The renal tissue level of α-SMA expression decreased and HGF expression significantly increased in Shenxikang group and other two related groups. Compared with those of the control group,the differences were statistically significant( P 〈0. 05). The improvement of those indexes of the Ziyinqingre group were significantly better than that of the Yiqihuoxue group( P 〈0. 05). The effective order of each treatment group was Shenxikang group Yiqihuoxue group Ziyinqingre group. Conclusion: Shenxikang can achieve significant medical effect in inhibit the expression of α-SMA in renal tissue and increase the level of HGF. One of the mechanisms may be the prevention of glomerulosclerosis and renal fibrosis.
