摘 要:目的:探讨三七总皂对实验性酒精性肝硬变大鼠肝纤维化治疗作用的分子机制。方法:选取健康成年雄性Wistar大鼠36只,使用体积分数50%酒精16 m L·(kg·d)^(-1)灌胃造模,9周后随机分成模型组、安珐特组、三七总皂苷组,每组12只。安珐特组给予西药安珐特21.6 mg·(kg·d)^(-1)进行灌胃,三七总皂苷组给予三七总皂苷100 mg·(kg·d)^(-1)进行灌胃,模型组继续使用体积分数50%酒精灌胃。同时设置空白组。12周后处死大鼠,剪下肝右叶,提取肝星状细胞总蛋白,测定各组蛋白含量,继而进行等电聚焦双向电泳,比较组间的差异蛋白点。结果:通过组间两两比较发现,组间的蛋白表达均存在着差异,正常组与模型组比较,共有23个差异点;正常组与安珐特组比较,共有33个差异点;正常组与三七总皂苷组比较,共有30个差异点;模型组与安珐特组比较,共有30个差异点;模型组与三七总皂苷组比较,共有31个差异点;安珐特组与三七总皂苷组比较共有30个差异点。结论:三七总皂苷对实验性酒精性肝硬变大鼠具有保护作用,其机制可能与三七总皂苷引起肝星状细胞蛋白质表达差异有关。Objective: To investigate the proteomics effect of Panax Notoginseng Saponinson of the hepatic stellate cells of rate with Alcoholic Cirrhosis to know more about the molecular mechanism of in medicating of the liver fibrosis in rats. Methods: 36 Healthy adult male Wistar rats were modeled by using the volume fraction of 50% alcohol 16 m L·( kg·d)^(-1)and were randomly divided into model group,AFT group and PNS group after 9 weeks( n = 12). Positive drug group was fed with western AFT21. 6 mg /( kg·d^(-1)),the PNS group was treated using 100 mg·( kg·d)^(-1)and the model group was treated by using the volume fraction of 50% ethanol gavage. Also a blank group was set. After 12 weeks,the rats were sacrificed; the total protein of the hepatic stellate cells of each group was extracted and measured. Two-dimensional electrophoresis of isoelectric focusing was conducted and differentially expressed proteins between the groups were compared. Results: the protein expressions between the groups were there different between the groups. There were a total of 23 points of difference. There were a total of 33 points of difference between the normal group and the AFT group. There were a total of 30 points of difference between the normal group and the PNS group. There were a total of 30 points of difference between the model group and the AFT group. There were a total of 31 points of difference between the model group and PNS group. There were a total of 30 points of difference between AFT group and PNS group. Conclusion: PNS plays a protective effect on rats with alcoholic cirrhosis,the mechanism of which may be differentially expressed proteins in hepatic stellate cells caused by PNS.
