摘  要:以不同封育年限恢复群落的大针茅和克氏针茅为研究对象,采用RAPD分子标记和核糖体rDNA的ITS序列分析方法,对大针茅和克氏针茅进行了遗传分化方面的研究.结果表明:(1)大针茅和克氏针茅亲缘关系较近又相对独立,且保持着一定的遗传差异.(2)不同恢复群落的大针茅、克氏针茅保持着较高的遗传多样性,12个引物共检测到292条扩增片段,其中245条具多态性,占到83.90%.(3)围栏封育过程中,两种针茅的遗传多样性发生了变化,而且在不同的围封样地间,大针茅和克氏针茅的遗传分化均是显著的(P<0.001),在总的遗传变异中占据着显著的比例,这表明围封在遗传分化中发挥了作用.The Stipagrandis and S.kryloviiin sample areas of different eclosuring periods were investigated.Genetic differentiation of S.grandis and S.krylovii were studied by RAPD analysis and phylogenetic analysis based on internal transcribed spacer(ITS).The results showed that:1)S.grandis and S.krylovii had a close genetic relationship but also keep certain genetic variability in sample areas,2)high genetic diversity exists in the S.grandis and S.krylovii in different restoring communities,since 292 amplified fragments were obtained by 12 pairs of primers,in which 245 were polymorphic(83.90%),3)during enclosuring,the genetic diversity of S.grandis and S.krylovii changed.Moreover,the genetic differentiation of S.grandis and S.krylovii were significant(P〈0.001)among different sample areas and accounted for significant proportion in the overall genetic differentiation.In conclusion,the enclosure treatment had a significant effect on the genetic differentiation of S.grandis and S.krylovii in different sample areas.
