摘 要:目的评价栀子大黄汤(Zhi-Zi-Da-Huang decoction,ZZDHD)抗酒精性肝损伤的药效作用。方法建立体外酒精性大鼠肝细胞损伤模型,考察ZZDHD含药血清对损伤肝细胞的增殖(methylthiazoletrazolium,MTT)及谷丙转氨酶(alanine aminotransferase,ALT)泄漏的影响。建立酒精性大鼠肝损伤体内模型,不同剂量给药后测定血清ALT,谷草转氨酶(aspartate aminotransferase,AST)和甘油三酯(triglyceride,TG)及肝组织中超氧化物歧化酶(superoxide dismutase,SOD),谷胱甘肽(reduced glutathione,GSH)和丙二醛(malondialdehyde,MDA)的含量与活性,并进行肝组织病理学检查。结果体外细胞试验中,体积分数为20%的含药血清能非常显著提高损伤肝细胞的增殖能力并降低细胞ALT泄漏(P<0.01),其活性最佳。大鼠体内,ZZDHD高、中剂量组均能不同程度地降低由酒精诱导的血清ALT、AST和TG水平的升高(P<0.01或P<0.05);均能非常显著性降低受损肝组织中的MDA含量(P<0.01),升高肝组织SOD活力和GSH含量(P<0.01),同时组织病理学也得出上述一致的结论。结论 ZZDHD对酒精性肝损伤具有明显的防治作用,其作用机制可能与减轻自由基和脂质过氧化物对肝细胞的损伤有关。Objective To investigate the effect of Zhi-Zi-Da-Huang Decoction( ZZDHD) on cells and rats with alcohol-induced hepatotoxicity. Methods The model of alcohol-induced hepatotoxicity was established by adding ethanol onto rat liver derived cell line( BRL-3A),and the efficacy of rat medicated serums on BRL-3A cells from alcoholic injury were evaluated by detecting the cell growth function( MTT methods) and ALT in the media. All rats except those in normal control group were administrated orally with 56% alcohol at 2 h after doses everyday for five consecutive days. The serum levels of ALT,AST activities and TG contents were measured. Moreover,the possible protective mechanisms were elucidated by assays of liver enzyme activities and component contents including SOD,GSH and MDA. The biochemical observations were supplemented by histopathological examination. Results All indexes of rats in the 20% drug containing serum group were shown to be significantly different( P〈0. 01) compared to model group,therefore 20%was chosen to be the optimum drug concentration. The high dose and middle dose of ZZDHD in rats exhibited significant and dose-dependent protective effect on alcohol-induced liver injury by reversing the increase of ALT and AST activities and TG content in serum( P〈0. 01 or P〈0. 05) decreasing the hepatic MDA level( P〈0. 01) and increasing the hepatic SOD and GSH activities( P〈0. 01). Meanwhile,the histopathology observation of the liver tissue was in accordance with above results. Conclusions ZZDHD exerts a hepatoprotective effect on alcohol-induced liver injury on cells and rats,and the mechanism of this activity is possibly related to its antioxidant properties.
