HPLC法同时测定桑叶药材及其炮制品中绿原酸、芦丁和异槲皮苷的含量

摘　　要：目的：建立同时测定桑叶药材及其炮制品炒桑叶、蜜桑叶中绿原酸、芦丁和异槲皮苷含量的方法。方法：采用高效液相色谱法。色谱柱为Agilent Zorbax SB-C18,流动相为乙腈-0.2%磷酸（梯度洗脱）,流速为1.0 ml/min,检测波长为350 nm,柱温为30℃,进样量为10μl。结果：绿原酸、芦丁、异槲皮苷的检测质量浓度线性范围分别为8.20～82.01μg/ml（r=0.999 9）、2.76～27.60μg/ml（r=0.999 9）、4.74～47.39μg/ml（r=0.999 9）;精密度、稳定性、重复性试验的RSD〈2%;加样回收率分别为98.58%～100.91%（RSD=1.02%,n=6）、99.19%～101.00%（RSD=0.82%,n=6）、98.41%～101.51%（RSD=1.08%,n=6）。结论：该方法操作简便,稳定性、重复性好,可用于桑叶药材及其炮制品中绿原酸、芦丁和异槲皮苷含量的同时测定。OBJECTIVE：To establish a method for the simultaneous contents determination of chlorogenic acid,rutin and isoquercitrin in Morus alba,fried M. alba and honeyed M. alba. METHODS：HPLC was performed on the column of Agilent Zorbax SB-C18 with mobile phase of acetonitrile-0.2% phosphoric acid（gradient elution）at a flow rate of 1.0 ml/min,detection wavelength was 350 nm,the column temperature was 30 ℃,and injection volume was 10 μl. RESULTS：The linear range was 8.20-82.01μg/ml for chlorogenic acid（r=0.999 9）,2.76-27.60 μg/ml for rutin（r=0.999 9）and 4.74-47.39 μg/ml for isoquercitrin（r=0.999 9）;RSDs of precision,stability and reproducibility tests were lower than 2%;recoveries were 98.58%-100.91%（RSD=1.02%,n=6）,99.19%-101.00%（RSD=0.82%,n=6）and 98.41%-101.51%（RSD=1.08%,n=6）,respectively. CONCLUSIONS：The method is simple with good stability and reproducibility,and can be used for the simultaneous determination of chlorogenic acid,rutin and isoquercitrin in different processed drugs of M. alba.
