摘  要:观察芒果苷对自发性高血压大鼠(SHR)炎症反应、单核细胞活化及血清炎性因子表达的影响,进一步了解芒果苷在抗高血压病慢性炎症中的作用及其机制。采用全自动无创血压测量系统进行尾动脉收缩压测定;采用细胞计数板进行全血白细胞计数;采用趋化实验计数外周血单个核细胞的趋化数;采用酶联免疫法测定大鼠血清中单核细胞趋化因子-1(MCP-1)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、白介素10(IL-10)的含量。给药前后3个时间点的模型组血压均较正常对照组显著升高(P<0.01),芒果苷各剂量组与模型组比较,血压有所降低,但差异没有统计学意义(P(0.05);芒果苷高剂量可明显降低模型组全血白细胞总数(P<0.05);芒果苷高、低剂量可明显降低模型组单核细胞趋化数(P<0.05);芒果苷各剂量均可明显降低模型组MCP-1含量(P<0.05);芒果苷低剂量可明显降低模型组大鼠TNF-a含量(P<0.05),芒果苷中、低剂量均显著升高模型组大鼠IL-10含量(P<0.01)。结果表明SHR存在明显的炎症反应,芒果苷对SHR没有明显的降压药效,但有明显的抗炎药效,其机制可能与抑制单核细胞的活化,调节炎性细胞因子表达有关。To study the effects of mangiferin on the inflammatory response, the activation of monocytes and the expression of inflammatory factors in serum of spontaneously hypertensive rats(SHR) ,to learn more about the role of mangiferin for resisting hypertension of chronic inflammation and its mechanism. Tail artery systolic pressure was measured by fully au- tomatic noninvasive blood pressure measurement system;The total numbers of whole blood leukocyte were counted by cell-count boards ; the chemotaxis numbers of peripheral blood mononuclear ceils were counted by chemotaxis test and the levels of monocyte chemotaxis factor-1 ( MCP-1 ), tumor necrosis factor alpha( TNF-a), interleukin 6 (IL-6) and interleu- kin 10 (IL-10)in rat serum were measured by using ELISA. In the three time points of before and after drug administra- tion, the blood pressure in model group was significantly higher than the normal control group(P 〈 0.01 ), and the blood pressure in each dose mangiferin group was lower than that in model group respectively, but with no statistical signifi- eance(P 〉0.05) ;High dose mangifcrin can significantly decrease the total numbers of whole blood leukocytes in the model group( P 〈 0.05 ) ;High and low dose mangiferin can significantly decrease the number of monocyte chemotaxis in the model group(P 〈0.05) ;Each dose of mangiferin can decrease the content of MCP-1 in the model group(P 〈0. 05); Low dose mangifefin significantly reduced the content of TNF-a in the model group(P 〈0.05) ;Medium and low dose man- giferin significantly increased the content of IL-10 in the model gmup(P 〈0.01 ). There was obvious inflammatory reaction in SHR,mangiferin had no obvious pharmacodyrmmics of hypotensive of SHR, but it bad obvious anti-inflammatory effect, its mechanism was possibly related to the inhibition of activation of monocvtes and the expression of inflammatory cytokines.
