摘  要:为明确引起掌叶半夏疫病的病原菌,本研究对河北省安国地区掌叶半夏疫病株进行了病原菌的分离纯化、致病性测定以及病原菌的形态学观察和rDNA-ITS序列鉴定。从发病植株上分离得到了疫霉菌、镰刀菌、链格孢菌等几种候选病原菌,健康植株在接种疫霉菌菌株后,出现了与田间病株相似的症状。该疫霉菌株最高生长温度为36℃,异宗配合,孢子囊有明显的一个乳突,脱落孢囊柄平均长度约为2.9μm,其rDNA-ITS序列比对结果与寄生疫霉(Phytophthora parasitica)同源性为99%。结合以上结果,最终确定引起掌叶半夏疫病的病原菌为Ph.parasitica。In order to know the pathogen causing blight disease on Pinelliapedatisecta,the pathogenicity,morphology characters,and rDNA-ITS of the pathogen isolated from the diseased P.pedatisecta grown in Anguo county were analyzed in the study.The results showed that some candidate pathogens were isolated,such as Phytophthora,Fusarium,and Alternaria.After inoculated with the Phytophthora sp.,the healthy P.pedatisecta showed symptoms similar to that observed in the field.The Phytophthorasp.is heterothallism mating type,the maximum growth temperature of which is 36℃,the sporangium has an obvious convex papillate,and the average length of caducous sporangiophore is about 2.9μm.BLAST analysis of the rDNA-ITS fragment sequence showed a 99% homology with the corresponding sequence of Ph.parasitica.Combination of the above results showed that the pathogen causing blight disease is Ph.parasitica.
