摘 要:目的探讨T细胞α型趋化因子(I-TAC)及CXC趋化因子受体3(CXCR3)在胃癌组织中的表达水平及临床意义。方法选取2013年1月至2016年1月本院普外科手术切除或黏膜活检所取的胃癌组织40例,相应的癌旁胃组织40例及正常胃黏膜组织20例,采用S-P免疫组织化学技术及逆转录聚合酶链反应技术分别检测I-TAC及CXCR3在三个研究组中的表达水平;并分析I-TAC及CXCR3mRNA的表达与常见临床病理参数的关系。结果胃癌组织中I-TAC及CXCR3的mRNA和蛋白质表达水平均高于癌旁胃组织及正常胃组织。免疫组化结果提示,I-TAC在胃癌组中阳性表达率(80.0%)高于癌旁组(52.5%)及正常胃组织组(55.0%),差异有统计学意义(P〈0.05);CXCR3在胃癌组中阳性表达率(67.5%)高于癌旁组(45.0%)及正常胃组织(40.0%),差异有统计学意义(P〈0.01)。临床病理因素分析提示,CXCR3的表达与胃癌组织的分化程度、淋巴结转移等呈正相关(P〈0.05)。结论胃癌组织中存在I-TAC及其受体CXCR3的特异性激活,提示I-TAC/CXCR3信号传导通路在胃癌发生及侵袭过程中发挥重要作用,可以作为一个诊断和治疗的分子靶点。Objective To investigate the expression and clinical significance of I-TAC and its receptor CXCR3 in gastric carcinoma tissue. Methods 40 cases of gastric cancer tissue, 40 cases of para-carcinoma tissue, and 20 cases of normal gastric tissues from patients undergoing exairesis or biopsy during January 2013 and January 2016 were collected. The expression of I-TAC, CXCR3 in surgical specimens were determined by reverse transcription-polymerase chain reaction and immunohistochemistry respectively. The association of I-TAC, CXCR3, and clinicopathological characteristics were analyzed. Results mRNA expression of I-TAC and CXCR3 in gastric cancer tissue were significantly higher than those in normal gastric tissue and para-carcinoma tissue. The positive expression rate of I-TAC in gastric cancer tissue, para-carcinoma tissue, and normal gastric tissue were 80.0%, 52.5%, 55.0% respectively, with statistically significant difference (P〈0.05). The positive expression rate of CXCR3 in gastric cancer tissue was 67.5%, significantly higher than 45.0% in para-carcinoma tissue and 40.0% in normal gastric tissue, with statistically significant difference (P〈0.01). The expression of CXCR3 in the tissueof gastric carcinoma was positively related to histologic differentiation, lymph node metastasis. Conclusion I-TAC and CXCR3 are highly activated in gastric cancer tissue, which indicates that I-TAC/CXCR3 signaling pathway may play a important role in the process of development and metastasis, so it may be used as a therapeutic target.
