摘 要:目的制备芦荟凝胶的胶原-壳聚糖-硫酸软骨素三元膜,研究其在大鼠皮肤深Ⅱ度烧伤创面愈合过程中的作用。方法制备芦荟凝胶的胶原-壳聚糖-硫酸软骨素三元膜。取大鼠60只,随机分为模型对照组、湿润烧伤膏组、三元膜组,每组20只。造成深Ⅱ度烧伤模型后,测量大鼠烧伤后皮肤创面平均愈合时间及第1、3、7、14天创面愈合率,检测烧伤后第1、14天皮肤创面组织中超氧化物歧化酶(SOD)和丙二醛(MDA)含量、血清中肿瘤坏死因子(TNF-α)含量并作统计学分析。结果与模型对照组相比,三元膜组能缩短烧伤大鼠皮肤创面愈合的时间(P<0.05),提高第3、7、14天的创面愈合率(P<0.01)。烧伤后第14天,与模型对照组相比,三元膜组皮肤创面组织中SOD含量升高(P<0.01),MDA含量降低(P<0.01),血清中TNF-α含量降低(P<0.01)。以上指标,三元膜组和烧伤膏组比较差异无统计学意义(P>0.05)。结论芦荟凝胶的胶原-壳聚糖-硫酸软骨素三元膜可通过清除氧自由基、减少促炎症因子的生成,从而促进大鼠皮肤深Ⅱ度烧伤的愈合。Objective To prepare the collagen-chitosan-chondroitin sulfate composite membrane with aloe vera gel,andinvestigate its effect in the healing process of deep Ⅱ partial thickness burn in rats. Methods The collagen- chitosan-chondroitin composite membrane with aloe vera gel were prepared. A total of 60 rats were randomly divided into 3 groups,thatis,model control group,moisture exposed burn ointment group,and composite membrane group,with 20 animals in each group.After deep Ⅱ partial thickness burn was inflicted,the rats were given corresponding treatment. The average skin wound healingtime after burn,and the wound healing rates in days 1,3,7 and 14 were observed and calculated. The contents of superoxidedismutase(SOD)and malondialdehyde(MDA)in skin wound tissue and the serum level of tumor necrosis factor-α(TNF-α)in days 1 and 14 were determined and then analyzed statistically. Results Compared with model control group,the compositemembrane group had obviously shorter shorten skin wound healing time(P〈0.05),and significantly higher wound healing ratesin days 3,7 and 14(P〈0.01). Compared with the rats of model control group in 14 d after burn,the content of SOD was elevated(P〈0.01)while that of MDA reduced(P〈0.01)in the skin wound tissue of composite membrane group,and the serum level ofTNF-α was decreased too(P〈0.01). There were no differences in composite membrane group and moisture ointment group.Conclusion The collagen-chitosan-chondroitin composite membrane with aloe vera gel can accelerate the healing process ofdeep Ⅱ partial thickness burn in rats by removing oxygen free radicals and reducing secretion of pro-inflammatory factor.
