摘  要:为了解副猪嗜血杆菌(HPS)OMPP2基因地方菌株的变异特点,本研究对2013年5月至2015年9月分离的11株四川地区HPSOMPP2基因进行序列分析。结果显示11株分离株与25株参考株的核苷酸同源性为93.5%~100%,核苷酸一致性低于90%,总体变异相对较大,尤其HPS-3、-8在OMPP2基因序列的第798~868位插入69个碱基,该变异在国内罕见,这与欧洲株和日本株的OMPP2基因在此位段的变异相似,进化树显示HPS-3和HPS-8株也与欧洲株和日本株亲缘关系最近,其中与Sw114日本株同源性最高(99.2%),因此推测该菌很有可能由日本传入,并且HPS-3和HPS-8株于不同年份从同一地区分离,据此推测HPS-3和HPS-8株很有可能是同一株菌,在该地区持续流行。本研究为四川地区HPS的基因突变和遗传变异研究提供参考资料。To investigate the variation feature of Haemophilus parasuis in Sichuan area, eleven isolates isolated from Sichuan region were analyzed based on outer membrane protein P2 (OMPP2). Sequence analysis displayed that the 11 isolates shared 93.5%-100% nucleotide homology with 25 reference strains, and the consistency was below 90%. Particularly, the isolates of H. parasuis-3 and H.parasuis-8 had a 69 bp-insertion at 798 bp-868 bp which was only found in foreign strain, such as C5 and D74 Sweden strain, Sw114 Japan strain, H465 Germany strain. The Phylogenetic tree shown that H.parasuis-3 and H.parasuis-8 had closer relationship with Europe and Japan strains, but shared 99.2% nucleotide homology with Swll4 Japan strain, so we speculated that the H.parasuis-3 and H.parasuis-8 were more likely to be introduced fi'om Japan, and both of H.parasuis-3 and H. parasuis-8 were isolated in different year but the same area, so they might be likely to originate from the same strain, and might be persistent prevalence in that areas. These data provide material for the study of variations of H.parasuis in Sichuan region.
