摘 要:为了解褐黄血蜱唾液内细菌的种类和传播途径,本研究分别提取褐黄血蜱饱血、半饱血的蜱虫及其唾液细菌总DNA,以其为模板,以通用引物扩增细菌16S r DNA V3区,并对其进行序列分析。结果表明,唾液内优势菌序列分别与狡诈球菌、鞘氨醇单胞菌属、立克次体、不可培养的细菌序列高度相似。虫体的优势菌序列与葡萄球菌属、立克次体、柯克斯体属的序列相似。饱血、半饱血蜱的唾液菌群结构存在一定差异,而蜱虫与其唾液的菌群结构差异很大。本研究表明褐黄血蜱唾液含条件致病菌,有经唾液传播疾病的潜在风险。To investigate the species and the transmission routes of bacterial communities in the saliva from adult female Haemaphysalis tlava, bacterial total genomic DNA was extracted from engorged and partly engorged tick bodies and their saliva from H.flava. Bacterial 16S rDNA V3 region were amplified with universal primers and sequenced. The results showed that sequences of dominant bacteria in the saliva were highly similar to sequences of Dolosicoccus paucivorans, Sphingomonas sp., Rickettsia peacockii and Uncultured bacterium, respectively. Sequences of dominant bacteria in tick bodies were similar to sequences of Staphylococcus sp., Rickettsia, Coxiella sp. The microbiota in the saliva had certain difference between engorged and partly engorged adult females, and the difference of the microbiota between tick bodies and their saliva was remarkable. The results indicate that the saliva from H.tlava contains opportunistic pathogen and has potential pathogenic risk.
