摘  要:为了解豫南地区猪场猪繁殖与呼吸综合征病毒(PRRSV)流行情况和流行毒株基因的遗传变异规律,本研究对2013年~2014年豫南地区部分发病猪场进行PRRSV检测,同时对分离株ORF5基因进行RT-PCR扩增、克隆和测序,并将其进行序列比对和遗传进化分析。结果表明:本研究检测的275份病料中,检测阳性样本为86份,阳性率为31.3%;测序的13株流行毒株,遗传关系上均属于美洲株;与VR2332毒株序列比对,13株分离株糖基化位点和抗原表位都有不同程度的变异;另外,遗传进化树分析表明,13株分离株中12株与2006年中国高致病PRRSV代表毒株JXA1同属一个亚群,1株与HB-1(sh)/2002毒株同源性较高,表明目前豫南地区以高致病性PRRSV毒株流行为主,同时还有少部分过渡性毒株存在。研究结果为豫南地区PRRS的防治及新型疫苗的研制提供了依据。To investigate the prevalence and the genetic variation of porcine respiratory syndrome virus(PRRSV)in southern region of Henan province,the virus was detected from swine clinical tissue samples from 2013 to 2014,ORF5 genes were amplified,cloned and sequenced for analyzing the genetic variation of PRRSV by reverse transcription polymerase chain reaction(RT-PCR).The results showed that 86 of 275tissue samples were positive,and positive rate was 31.3%;The homologies of ORF5 genes among the 13 PRRSV isolates were 95.2%~99.7% and 93.0% ~99.5%in nucleotide and amino acid sequences,respectively;And 13 PRRSV isolates belonged to Northern American genotype.Compared with VR2332 strain,some variations were found in the glycosylation sites and antigen epitope of the 13 ORF5 genes.Furthermore,the phylegenetic tree of ORF5 of 13PRRSV isolates indicated that 12 PRRSV isolates were in the same subgroup with the highly pathogenic PRRSV emerged in south China in 2006,and 1PRRSV isolate was loosely related to the reference strain HB-1(sh)/2002.The result showed that the highly pathogenic PRRSV had become the dominant PRRSV epidemic strain in southern region of Henan province,but intermediate strain still exists in an circulate way.This study would provide a theoretical basis for the disease prevention and development of new vaccine of PRRSV in southern region of Henan Province.
