摘 要:目的探讨山茱萸环烯醚萜苷类特征成分马钱苷、莫诺苷对糖基化终末产物(AGES)诱导人脐静脉内皮细胞(HUVEC)损伤的保护作用.方法体外培养HUVEC,用马钱苷、莫诺苷(终浓度分别为100、10、μmol·L^-1)预孵1h后,再加人AGE/(200mg·L^-1)刺激,并设氨基胍为阳性对照,孵育24h后,采用MTT法检测马钱苷、莫诺苷对HUVEC增殖率的影响;采用试剂盒检测细胞上清液中-氧化氮(NO)、内皮素(ET-1)、单核细胞趋化蛋白-1(MCP-1)和血管细胞黏附分子-1(VCAM-1);采用Westernblot法检测HU-VEC中糖基化终末产物受体(RAGE)、核转录因子(NF-kB)的蛋白表达.结果山茱萸环烯醚萜苷类特征成分马钱苷、莫诺苷能抑制AGEs导致的HUVEC损伤,与空白对照组比较,模型组细胞RAGE、NF-kB蛋白表达上调(/3〈0.01),ET-1、MCP-1、VCAM-1分泌增加(P〈0.01),NO水平降低(P〈0.01).马钱苷、莫诺苷给药组能不同程度地下调RAGE、NF-kB蛋白表达,减少ET-1、MCP-1、VCAM-1分泌,升高NO水平,与模型组比较差异具有显著性(P〈0.05-0.01).结论山茱萸环烯醚萜苷类特征成分马钱苷、莫诺苷对AGEs导致的HUVEC损伤具有明显的保护作用,其作用机制为下调RAGE蛋白表达,抑制RAGE受体相关NF-κB信号通路发挥抗炎作用,从而改善内皮细胞功能.To observe the protective mechanismof loganinand morroniside ( active components in Cornnsofficinalis) on HUVEC injury induced by advancedglycation end products ( AGEs ). Methods HUVEC swere cultured in vitro and divided into control group,model group ( AGEs group ) , loganin group, morronisidegroup and aminoguanidine group ( set as positivecontrol) . After being incubated with loganin and morroniside( final concentrations were 100,10,1 μmol·L^-1) for 1 h , HUVEC s were stimulated by AGEs of200 mg·L^-1 for 24 h . Then , the cell viability wasmeasured by using M T T method. The supernatant wasextracted and the levels of N O , ET-1 , MCP-1 , VCAM-1were measured by the corresponding kits. Receptors ofadvanced glycation end products ( RAGE ) and N F -k Bin HUVEC were detected by Western blot. ResultsLoganin and morroniside could inhibit HUVEC injuryinduced by AGEs . In model group,the contents of ET-1,MCP-1,VCAM-1 increased (P 〈0 .01),the contentof N O decreased (P〈0.01) and the expression ofRAGE and NF-k B increased(P〈0. 01) ; however, loganinand morronside could reduce the ET-1, MCP-1,V CAM-1 contents, increase the NO content and downregulatethe expression of RAGE and N F-k B to differentextents. Conclusion Loganin and morronisidecould ameliorate HUVEC injury,and its mechanismmay be related to inhibit inflammation, the improvementof endothelial cell function , and the decrease ofthe expression of RAGE .
