摘  要:目的了解山东省不同地理种群淡色库蚊对拟除虫菊酯类杀虫剂的抗药性水平和kdr等位基因的频率分布,探讨不同地理种群淡色库蚊kdr等位基因频率同抗药性水平之间的相关性。方法采用WHO推荐浸渍法检测蚊虫抗药性,采用AS·PCR扩增技术检测成蚊的kdr等位基因频率,利用线性回归方法分析kdr等位基因频率与抗药性的相关性。结果济南、淄博、济宁3个地理种群淡色库蚊对溴氰菊酯均产生抗药性,抗性系数分别是4.14、5.29和2.86;淄博种群对高效氯氰菊酯产生抗药性,抗性系数为48.68,济南种群对氯氰菊酯产生抗药性,抗性系数为4.36。济南、淄博、烟台、济宁种群淡色库蚊的kdr频率(%)分别是是44.71%、54.22%、21.84%、32.97%,淄博种群最高,其次是济南种群。不同种群的溴氰菊酯抗性水平与抗性等位基因频率有显著的相关性(r2=0.981 6,P=0.009 2),高效氯氰菊酯和氯菊酯抗性水平与抗性等位基因频率无显著的相关性(r2分别为0.590 4和0.020 1,P值分别为0.231 6和0.858 2)。结论山东省4个不同地理种群淡色库蚊对拟除虫菊酯类药物的抗性水平由高到底依次为淄博种群、济南种群、济宁种群、烟台种群,对3种药物都有不同程度的抗药性产生,其中溴氰菊酯抗性水平最高,其次是高效氯氰菊酯和氯氰菊酯;野外种群淡色库蚊对溴氰菊酯抗性水平与kdr等位基因频率呈显著相关性,高效氯氰菊酯、氯氰菊酯抗性水平与kdr等位基因频率无显著相关性。Objective To investigate the resistances of Culex pipiens paUens from four geographical populations in Shandong Province to deltamethrin, β - cypermethrin, permethrin and the frequencies of kdr alleles in every population, to discuss the correlation between Kdr allele frequencies with resistance levels of Culex pipiens pallens among different geographical populations. Methods The resistances were determined using the dipping method recommended by WHO and the frequencies of kdr allele in different populations of adult Culex pipiens pallens were determined by allele - specific PCR ( AS - PCR). Linear regression analysis was used to determine the relationship between the frequencies of the kdr alleles and insecticide resistances. Results Different resistance levels to deltamethrin have been developed in Jinan,Zibo and Jining geographical populations of Culex pipiens pallens. The resistance ratios (RR) of three feild populations were 4.14,5.29 and 2.86 respectively. The RR of Zibo strains to beta - cypermethrin was 48.68 ,The RR of Jinan strains to permethrin was 4.36 ,the frequencies of kdr allele in Jinan strains,Zibo strains,Yantai strains and Jining strains were 44.71% , 54.22% , 21.84% and 32.97% respectively. The results showed that the kdr allele frequency significantly correlated with levels of resistance to deltamethrin among the population ( r2 = 0.9816, P = 0. 0092 〈 0.05 ), whereas it did not correlate with levels of resistance to [3 - cypermethrin ( r2 = 0. 5904, P = 0.2316 〉0.05) and permethrin (r2 =0.0201,P=0.8582〉0.05).Conclusion The results demonstrated that the pyrethroid resistance in Culex pipiens pallens from different geographical populations within Shandong Province in descending order was Zibo 〉 Jinan 〉 Jining 〉 Yantai. The four geographic populations of Culex pipiens pallens generally had different levels of resistance to deltamethrin, beta - cypermethrin and permethrin.
