摘 要:目的:探讨雷公藤甲素体外对人表皮干细胞增殖的影响。方法:应用中性蛋白酶-Ⅳ型胶原黏附方法分离正常人的包皮表皮干细胞,采用细胞免疫组化法检测培养传代细胞的角蛋白19(CK19)、角蛋白10(CK10)、β1整合素表达,鉴定人表皮干细胞。培养传代细胞中加入不同浓度的雷公藤甲素,用倒置显微培养后观察细胞的形态学变化,应用四甲基偶氮唑盐(MTT)法检测细胞的增殖情况,探索雷公藤甲素对人表皮干细胞增殖的影响。结果:免疫组化法检测提示培养传代细胞,β1整合素、CK19表达阳性,CK10表达阴性,MTT法检显示雷公藤甲素能显著抑制人表皮干细胞的增殖,且存在一定的浓度依赖性。结论:雷公藤甲素能有效抑制人表皮干细胞增殖。Objective: To investigate the effect of Triptolide on the proliferation of human epidermal stem cells ( hESCs ) in vitro.Method: hESCs was isolated from human foreskin by the method of protease-collagen Ⅳ-coated adhesion.The expression of integrins β1, keratin 19 and keratin 10 on the generation of hESCs were examined by immunocytochemical method.After the generation of hESCs were cultured in epilife medium containing Triptolide of different concentrations respectively.The shape, clone formation and proliferation of the cell were observed through inverted microscope.The effect of Triptolide was evaluate by MTT method on proliferation of human epidermal stem ceils in vitro.Result: The results of immunocytochemistry showed keratin 19 and intergrin β 1 were positively expressed at the generation of hESCs, but keratin l0 was negative.MTT assay showed that the proliferation of human epidermal stem cells was significantly inhibited, and there was a concentration dependent.Conclusion: Triptolide can effectively inhibit the proliferation of human epidermal stem cells in vitro.
