摘  要:目的:研究栝楼薤白半夏汤预处理对缺血再灌注大鼠心肌组织的保护作用及其机制是否与PI3K/Akt信号通路相关。方法:40只SD大鼠按照随机数字表法分为假手术组、模型组、预处理组、栝楼组。模型组及栝楼组大鼠行结扎冠状动脉左前降支30 min、再灌注2 h的方法造模,并设预处理组为对照。预处理组造模方法为短暂缺血5 min、再灌注5 min,反复3次后行缺血30min、再灌注120 min。栝楼组给予中药汤剂栝楼薤白半夏汤灌胃,其余各组灌胃同体积的生理盐水。生化自动仪检测大鼠血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、c Tn T(肌钙蛋白T)含量;采用免疫组化法检测心肌凋亡相关蛋白bax、bcl-2表达情况,采用免疫印迹法检测心肌组织的Akt、p-Akt表达。结果:栝楼组和预处理组血清CK-MB、LDH、c Tn T含量及心肌组织的bax蛋白表达低于模型组,而bcl-2、p-Akt蛋白表达高于模型组。结论:栝楼薤白半夏汤预处理对心肌缺血再灌注损伤具有保护作用,通过激活PI3K/Akt信号通路,与调节凋亡相关基因bcl-2、bax蛋白的表达有关。Objective: To study the effects of Gualou Xiebai Banxia Decoction on rats with myocardial ischemia /reperfusion injury and PI3 K / Akt mechanism. Methods: Forty male SD rats were randomly divided into sham operation group,model( ischemia reperfusion) group,ischemia preconditioning group,Gualou Xiebai Banxia decoction( Gualou)group. The model group were established by ligation of LAD 30 minutes and 120 minutes reperfusion. Gualou group were established by ischemia reperfusion model and previous three 5 minutes ischemia and 5 minutes reperfusion. Creatine kinase MB Isoenzyme( CK-MB),lactate dehydrogenase( LDH) and Troponin T( c Tn T) in serum were measured. The expression of the bax and bcl-2 was tested by immunohistochemistry method. The expression of Akt was tested by Western blot method.Results: The serum level of CK-MB,LDH,c Tn T and bax in myocardium of ischemia preconditioning and Gua Lou group were lower than model group,but higher than sham group. The expression level of Akt and bcl-2 in myocardium ischemia preconditioning and Gua Lou group were higher than IR group. Conclusion: Gualou Xiebai Banxia decoction preconditioning can protect ischemia reperfusion rats by uploading the expression of bcl-2 and download the expression of bax via PI3 K / Akt signal pathway.
