摘  要:目的:筛选地黄microRNAs(miRNAs)类药效物质及其靶基因功能分析。方法:采用高通量测序法,测定地黄、空白小鼠和喂食地黄匀浆液小鼠肝组织及血浆中miRNAs的表达丰度,并与Gene Bank上的植物microRNA数据库比对,结合荧光定量PCR验证,鉴定吸收进入体内的地黄miRNAs,以人类mRNA为靶标,采用Target Scan与MiRanda预测地黄miRNAs的靶基因,并通过GO与KEGG分析靶基因的相关功能。结果:共鉴定出2个地黄miRNAs可吸收进入体内,其主要在生化过程、细胞成分和分子功能3大区域发挥作用,并影响肿瘤、代谢、炎症等相关信号通路。结论:miRNAs可能是一类新型的中药药效物质。Objective: To investigate pharmacodynamic microRNAs( miRNAs) in Rehmannia glutinosa( R.glutinosa) and their target genes' functions. Methods: MiRNA profiles of R. glutinosa,in liver and plasma of control mice and R. glutinosa extract- treated mice were detected using next generation sequencing,and then the miRNAs which took into the organism were validated by Q- PCR using plant miRNA database in GeneBank. Human mRNA database as targets was used to predicte target genes of miRNAs by Target Scan and MiRanda analysis,while the target gene functions were considered by GO and KEGG analysis. Results: Two pharmacodynamic microRNAs were found and took part in biological processes,cellular components,molecular functions,which affected tumor,metabolism and inflammation- related signal pathways. Conclusion: MiRNAs might act as new pharmacodynamic materials of traditional Chinese medicine.
