摘  要:目的探讨肾上腺素β2受体(AR-β2)对失血性休克(HS)大鼠骨骼肌过氧化物酶体增殖物激活受体γ协同刺激因子(PGC-1)表达的影响。方法将62只SD大鼠随机分为对照组(n=6)、实验组(n=28)和干预组(n=28)。按22.5 ml/kg放血建立HS模型。干预组放血后,立即给予ICI 118.551以0.5 mg/kg静脉注射;休克30 min后,回输放出的血和2倍放血量的林格氏液;复苏期维持30 min,监测血流动力学8 h。记录建模前及建模后不同时点的红细胞比容(Hct)、乳酸、剩余碱和血糖。采用蛋白印迹法及定量聚合酶链反应(PCR)检测不同时间骨骼肌组织PGC-1蛋白及mRNA表达的变化。结果休克模型建立后,血乳酸升高、剩余碱减少。与实验组比较,干预组可以显著降低血乳酸并增加碱剩余。休克组动物的死亡率明显高于干预组。虽然复苏后2 h,实验组、干预组均出现PGC-1蛋白和mRNA的表达增加,但实验组增加更为显著,差异有统计学意义(P<0.05);其后,两组PGC-1蛋白和mRNA的表达均逐渐下降,两组差异无统计学意义(P>0.05)。结论 HS时,AR-β2可对PGC-1基因表达进行适应性调节,进而调控能量代谢平衡。Objective To investigate the effects of β2-adrenergic receptor on expressions of PGC-1 in skeletal muscles in rat model of hemorrhagic shock. Methods ds SD rats were randomized into three groups: Control( C),Hemorrhage( H) and Hemorrhage + AR-β2( N). Venous blood( 22. 5 ml/kg) was continously withdrawn over 10 minutes to establish hemorrhagic shock model. Selective β2-adrenoceptor antagonist ICI 118,551( 0. 5 mg / kg) was intravenously administered after hemorrhage had finished. After 30 minutes shock periods,rats received whole blood and lactated Ringer' s solution resuscitation( 1: 2) for 30 minutes. Rats were monitored for 8h after resuscitation. Hematocrit( Hct),Base excess( BE),Lactate( Lac) and Glucose( Glu) were recorded at the baseline and different hemorrhagic shock time( HST). The expressions of PGC-1 mRNA and protein in skeletal muscles were tested by real-time reverse transcription polymerase chain reaction( qRT-PCR) and western blotting. Results Rats in Group N had lower lactate levels and higher BE levels than Group H( p〈0. 05). At 72 hours,there were five survivors in Group H and eight in Group N( p〈0. 05 versus Group H,Log Rank). Both Group H and Group N increased PGC-1 mRNA and protein expressions at 2 hours after resuscitation,but more higher expressions were seen in Group H compared with Group N( p〈0. 05). Thereafter,the expressions of PGC-1 mRNA and protein decreased gradually in both groups,and no statistic differences were found between the two groups. Conclusion Adrenergic receptor-β2could regulate energy metabolism balance through adaptive regulation to PGC-1 during hemorrhagic shock.
