鸦胆子油交联环糊精包合物对结肠癌移植瘤生长的影响

摘　　要：目的：探讨鸦胆子油交联环糊精包合物（BJO-CDP）对小鼠结肠癌细胞CT-26移植瘤生长的影响,并与市售鸦胆子油乳剂（BJOE）抑瘤效果进行比较。方法：小鼠结肠癌细胞CT-26细胞株接种于4~6周龄雄性Balb/c小鼠的腋下,接种24 h后将60只小鼠随机分为5组,即对照组（生理盐水,0.03 m L/只）,BJOE高剂量组（180mg/kg）,BJO-CDP低剂量组（60 mg/kg）,BJO-CDP中剂量组（120 mg/kg）,BJO-CDP高剂量组（180 mg/kg）,灌胃给药。给药10 d。停药次日,称小鼠体质量,处死,称瘤质,计算抑瘤率。结果：给药后,BJOE高剂量组瘤质低于对照组,差异有统计学意义（P〈0.01）;BJO-CDP低、中、高剂量组瘤质均低于对照组,差异均有统计学意义（P〈0.05或P〈0.01）。BJO-CDP高剂量组瘤质低于BJOE高剂量组,差异有统计学意义（P〈0.05）。BJO-CDP高剂量组对小鼠体质量影响低于BJOE高剂量组,差异有统计学意义（P〈0.05）。结论：制备的鸦胆子油包合物能提高鸦胆子油抑制结肠癌细胞的能力。Objective： To investigate the effects of Brucea javanica Oil and β-cyclodextrin polymer（BJOCDP） inclusion complexes on mice colon CT-26 cell line in Balb/c mice of subcutaneous xenotransplanted tumor, and the commercial emulsion of BJO（BJOE） was used as a reference. Methods： Mice CT-26 colon cell was injected into Balb/c mice layers of armpit. 24 hours later, the 60 mice were randomly divided into 5groups： control group（physiological saline, 0.03 m L/mice）, BJOE high dose group（180 mg/kg）, BJO-CDP low dose group（60 mg/kg）, BJO-CDP medium dose group（120 mg/kg）, BJO-CDP high dose group（180 mg/kg）. Administered once daily for 10 days by gavage, in the 11 th day, mice were weighed, sacrificed, and tumors were excised for being weighed immediately, calculated tumor inhibition ratio. Results： Compared with control group,significant inhibition effects on colon cancer weight was observed in BJOE high dose group（P〈0.01）, and BJOCDP groups with low dose（P〈0.05）, middle dose（P〈0.01） and high dose（P〈0.01） were lower than control group.Moreover, the BJO-CDP groups had less colon cancer weight and weight loss of mice than BJOE groups（P〈0.05）.Conclusion： BJO-CDP can improve the ability of Brucea Javanica Oil inhibiting colon cancer cell.
