摘  要:目的观察三磷酸腺苷(ATP)敏感性钾通道(KATP)对福马林致炎症痛大鼠脊髓背角神经元神经激肽1受体(NKlR)内在化的影响。方法鞘内置管成功的雄性大鼠20只,大鼠随机分为4组(n=5):福马林炎性痛模型对照组(F组)、福马林炎性痛模型+鞘内注射吡那地尔(P组)、福马林炎性痛模型+格列本脲50μg/10μl(G组)、空白对照组(C组)。其中F、P和G组15只大鼠右后足掌皮下注射5%福马林液50μl即制作福马林炎性痛模型,空白对照组注射生理50μl。福马林或者生理盐水右侧后肢足掌皮下后30min行鞘内注射,F和C组生理盐水10ILl,P组10μg吡那地尔10μl,G组50斗g格列本脲10μl。行腰段脊髓NKlR免疫荧光染色;在荧光显微镜下观察统计,神经元胞体中内在化荧光颗粒≥10者为内在化阳性神经元。结果空白对照组腰段脊髓背角浅层内在化阳性神经元占神经元总数百分比[(20.3±3.5)%]明显低于F、P和G组,F、P和G组NK1R内在化分别为(91.2±2.3)%、(87.2±1.9)%、(95.1±1.7)%;与F组比较,P组鞘内注射10μg吡那地尔可以降低NKIR内在化比率,差异有统计学意义;G组鞘内注射格列本脲50μg可以显著增加NKIR内在化比率。结论改变KATP通道活动状态可能影响炎性痛的传导通路,此通道可能成为治疗此类疼痛治疗的靶点。Objective To investigate the effects of adenosine triphosphate (ATP) sensitive potas- sium channel (KATP) on intemalization of neurokinin 1 (NK1) receptor in dorsal horn of rat spinal cord. Methods ( 1 ) The rats were implanted with intrathecal catheters, 20 neurological normal rats were divid- ed into four groups (5 each): C group (control group, both intrathecal and subcutaneously injection of normal saline), F group (intrathecal normal saline with subcutaneously formalin), P Group (intrathecal 10 μg pinacidil and subcutaneously formalin) , G group (intrathecal 50 μg glibenclamide and subcutane- ously formalin). (2) Immmunohistochemistry was done after spinal cord was fixed by perfusion by forma- lin, and comparison was done between groups. Results Subcutaneously injection of formalin can induced internalization rate of NK1 receptor in rat spinal cord significantly(control is (20. 3 ±3.5)%, while F, P and G group were (91.2 ±2. 3)%, (87.2 ± 1.9)%, (95.1± 1.7)%, respectively. ), intrathecal injec- tion of pinacidil is able to decrease the internalization rate of NK1, while as, intrathecal injection glib- enclamide increases the internalization rate of NK1. Conclusion Manipulation of ATP-sensitive potassium channel can affect the internalization of NK1 receptor induced by paws formalin injection in rat. It indicates that neuronal ATP-sensitive potassium channel might play a role in the pain sensation.
