富血小板血浆对痤疮动物模型治疗作用的实验研究

摘　　要：目的建立兔耳痤疮动物模型,观察富血小板血浆(PRP)对兔耳痤疮动物模型的治疗效果。方法用油酸和痤疮丙酸杆菌菌液建立新西兰大白兔兔耳痤疮动物模型;将造模成功的新西兰大白兔分为分为PRP治疗组、生理盐水组和异维A酸乳膏阳性对照组(7只/组),于各组兔耳痤疮皮损处外涂PRP、生理盐水和异维A酸乳膏,1次/d,连续治疗14 d,取兔耳痤疮处皮肤组织活检,镜下观察病理改变。结果外用油酸涂抹和痤疮丙酸杆菌菌液注射14 d后,兔耳皮肤外观和组织学观察呈痤疮样改变,与人类痤疮相似;3组动物治疗14 d,根据疗效组织病理学判定标准对结果判定:PRP组3例痊愈,4例明显好转;异维A酸乳膏组5例痊愈,2例明显好转;生理盐水组1例明显好转、5例好转、1例无效(P<0.01)。结论 PRP对兔耳痤疮动物模型具有良好的治疗作用,与异维A酸乳膏有着相近的治疗效果。Objective To investigate the therapeutical effect of platelet rich plasma（ PRP） on the rabbit ear acne model. Methods The rabbit ear acne model was induced by oleic acid and propionibacterium acne bacteria liquid. Rabbits with ear acne were divided into three groups,with each group having seven animals： the PRP-treated group,the negative normal control group treated with saline group and the positive control group treated with Vitamin A acid cream. PRP,physiological saline and Vitamin A acid cream were applied to the acne lesions of the rabbits＇ ear in each group once a day for a total of 14 days. Skin tissue specimens were harvested from the same acne lesions of the rabbits＇ ear for biopsy. The pathological changes were observed under microscope. Results The application of oleic acid and injected propionibacterium acne bacteria liquid for 14 days led to acne changes in the skin of rabbits＇ ear,which were similar to the acne of human. After treatment for14 days,a statistical analysis was performed according to the effect of histopathological decision criteria. In the PRP group,3 recovered,4 improved notably; in the positive control group,5 recovered and 2 improved significantly; in the negative control group,1 improved notably,5 cases improved and 1 invalid was found. These results showed that there were significant difference between PRP-treated group and the negative control group（ P〈0. 01）. Moreover,no difference was found between PRP-treated group and the positive control group（ P〉0. 05）. Conclusion PRP has a good therapeutical effect on rabbit ear acne,which was similar to the effect of Vitamin A acid cream,a positive therapeutic drug.
