摘  要:目的:探讨炎症因子对人颗粒细胞凋亡调控基因P53、Bax、Casepase-3、Bcl-2、Survivin表达的影响。方法:选择行体外受精-胚胎移植(IVF-ET)治疗的非多囊卵巢综合征(PCOS)患者50例,提取卵泡液中颗粒细胞进行体外培养,随机分为炎症因子处理组和对照组,处理组分别加入5nmol/L白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNFα),比较各组颗粒细胞的增殖速率及凋亡调控基因的表达是否存在差异。结果:IL-1、IL-6、TNFα处理组体外培养48小时后颗粒细胞增殖速率低于对照组(P<0.05);处理组促凋亡基因P53、Bax、Casepase-3的表达水平高于对照组(P<0.05),而抗凋亡基因Bcl-2、Survivin的表达水平低于对照组(P<0.05)。结论:炎症因子IL-1、IL-6、TNFα抑制人颗粒细胞增殖,并诱发颗粒细胞凋亡。Objective: To explore the effect of inflammatory markers on the expressions of P53,bax,casepase-3,bcl-2 and survivin in human granulosa cells. Methods: Granulosa cells in follicular fluid were extracted from 50 cases of patients treated with IVF-ET without PCOS and cultured in vitro. Cells in experimental group were,respectively,treated with IL-1,IL-6 and TNF-α. The untreated cells were defined as control. The growth,proliferation rate and expression level of apoptosis-regulating genes were compared between the treatment and control group. Results: The proliferation rate of granulosa cells in the treatment group stimulated by IL-1,IL-6 and TNF-α were significantly lower than those in control group( P〈0. 05),after 48 h stimulation. The expression of pro-apoptotic gene including Bax,Casepase-3 and P53 in treatment group was higher than those in control group( P〈0. 05),while the expression of anti-apoptotic gene including Bcl-2 and Survivin was lower than those in control group( P〈0. 05).Conclusions: Inflammatory markers including IL-1,IL-6 and TNF-α can decrease the proliferation rate and induce the apoptosis in human granulosa cells.
