摘  要:目的:建立同时测定安喘舒丸中腺苷、黄芪甲苷含量的方法。方法:采用液质联用技术,以甲醇-0.1%甲酸溶液为流动相等度洗脱,以Agilent ZORBAX SB-C_(18)色谱柱为分析柱,通过电喷雾离子源,选择正离子模式多反应监测方式检测。结果:腺苷、黄芪甲苷的线性范围分别为10~320 ng·m L^(-1)(r=0.996 8)、5~160 ng·m L^(-1)(r=0.995 5),加样回收率在96.17%~99.48%,RSD均低于1.14%。结论:该法专属性强、快速灵敏,可用于安喘舒丸中腺苷、黄芪甲苷的含量测定。Objective: To develop a UPLC-MS/MS method for the determination of adenosine and astragaloside IV in Anchuanshu Pill. Methods: Isocratic elution was carried out with mobile phase consisting of methanol - 0. 1% formic acid. The separation was performed on an Agilent ZORBAX SB-C18 column, and the mass spectrometer was operated in the positive ionization electrospray (ESI) mode using multiple monitoring (MRM) for analysis of two components. Results: Adenosine and astragaloside IV were all determined exactly, the linear ranges were 10-320 ng. mL -1 ( r = 0. 996 8 ), 5-160 ng·mL^-1 ( r = 0. 995 5 ) , respectively. The recoveries of two analytes ranged from 96. 17%-99. 48% and the relative standard deviations were all below 1.14%. Conclusion: A sensitive, accuracy and suitable UPLC-MS/MS method has been developed, and the method could be applied for the determination of adenosine and astragaloside IV in Anchuanshu Pill.
