摘  要:目的:观察强骨饮对去卵巢骨质疏松大鼠骨显微结构的影响。方法:将36只10周龄雌性SD大鼠随机分为假手术组、强骨饮组和模型组,每组12只。假手术组切除卵巢周围脂肪,其他两组均切除双侧卵巢。造模后3个月,假手术组以生理盐水0.041 m L·g-1灌胃;强骨饮组以强骨饮浓缩液0.041 m L·g-1灌胃;模型组以碳酸钙D3混悬液0.01 m L·g-1、阿法骨化醇溶液0.037 m L·g-1灌胃;每日1次,连续干预2个月。药物干预结束后,采用micro-CT扫描大鼠股骨头至股骨中段,对图像进行三维重建,观察大鼠骨显微结构变化,并对3组大鼠各项骨显微结构参数进行比较。结果:药物干预2个月后,假手术组骨组织内骨小梁结构排列紧密、有序,骨小梁分离度低;强骨饮组和模型组骨小梁数量明显减少,排列稀疏或出现断裂,形态扭曲。3组间骨表面积(bone surface,BS)、骨体积(bone volume,BV)、骨表面积和体积比值(bone surface/bone volume,BS/BV)、骨体积分数(bone volume/tissue volume,BV/TV)、骨小梁厚度(trabecular thickness,Tb.Th)、骨小梁数量(trabecular number,Tb.N)、骨小梁分离度(trabecular separation,Tb.Sp)比较,差异均有统计学意义(F=63.243,P=0.000;F=10.290,P=0.000;F=43.774,P=0.000;F=10.685,P=0.044;F=10.489,P=0.000;F=140.827,P=0.000;F=382.812,P=0.000);强骨饮组和模型组BS、BV、BS/BV、BV/TV、Tb.Th、Tb.N均小于假手术组(P=0.000,P=0.018,P=0.000,P=0.045,P=0.004,P=0.037;P=0.000,P=0.008,P=0.000,P=0.031,P=0.004,P=0.000),Tb.Sp均大于假手术组(P=0.000;P=0.000);强骨饮组BS、BV、BS/BV、Tb.Th大于模型组(P=0.009,P=0.007,P=0.045,P=0.000),Tb.Sp小于模型组(P=0.000),2组BV/TV和Tb.N的组间差异均无统计学意义(P=0.104,P=0.869)。结论:与碳酸钙D3和阿法骨化醇联用相比,强骨饮更有利于改善去卵巢骨质疏松大鼠的骨显微结构,具有更好的抗骨质疏松作用。Objective: To observe the effect of Qianggu Yin( 强骨饮,QGY) on bone microstructure in ovariectomized osteoporosis rats.Methods: Thirty-six 10-week-old female SD rats were randomly divided into sham-operated group,QGY group and model group,12 cases in each group. The lipectomy around the ovary were performed on rats in sham-operated group,while the bilateral ovariectomy were performed on rats in QGY group and model group. The rats in sham-operated group were intragastric administrated with normal saline( 0. 041 ml / g),and the rats in QGY group were intragastric administrated with QGY concentrated solution( 0. 041 ml / g),while the others in model group were intragastric administrated with calcium carbonate D3 suspensions( 0. 01 ml / g) and alfacalcidol solution( 0. 037 ml / g) at 3 months after the modeling,once a day for consecutive 2 months. After the end of drug intervention,the femoral head and middle segment of femur of the rats were scaned by micro-CT,and the images were inputed into the computer to build three-dimensional reconstructed images. Then the bone microstructure changes in rats were observed and the bone microstructure parameters were compared between the 3 groups. Results:After 2-month drug intervention,compact and orderly bone trabecula were found in the bone tissue in sham-operated group with low separation degree,while sparse or broken bone trabecula were found in QGY group and model group with distorted shape. There was statistical difference in bone surface( BS),bone volume( BV),ratio of bone surface to bone volume( BS / BV),bone volume fraction( BV / TV),trabecular thickness( Tb. Th),trabecular number( Tb. N) and trabecular separation( Tb. Sp) between the 3 groups( F = 63. 243,P = 0. 000; F =10. 290,P = 0. 000; F = 43. 774,P = 0. 000; F = 10. 685,P = 0. 044; F = 10. 489,P = 0. 000; F = 140. 827,P = 0. 000; F = 382. 812,P =0. 000).
