摘  要:为了探讨中哈边境阿拉山口口岸地区凶小库蚊核糖体DNA第二转录间隔区(r DNA-ITS2)基因序列特征,分析其遗传变异和亲缘关系,为口岸地区蚊的鉴定及疾病防治奠定基础。本研究提取中哈边境阿拉山口口岸地区凶小库蚊蚊虫DNA,PCR扩增凶小库蚊的r DNA-ITS2基因并测序。将测序结果进行Blast分析,使用Mage 6.0构建分子进化树。结果显示:PCR扩增获得凶小库蚊第二转录间隔区(r DNA-ITS2)323 bp左右的基因片段。Blast显示与Culex restuans(U22136)具有较高的同源性,与其他库蚊属蚊种序列间差异集中在24-323 bp,差异表现为插入/缺失、转换/颠换、简单重复单元拷贝数不同等。结论:本研究报道了凶小库蚊第二转录间隔区(r DNA-ITS2)基因序。阿拉山口口岸凶小库蚊第二转录间隔区(r DNA-ITS2)基因区显示存在遗传多态性,可能与蚊虫地理区域,生态环境和贸易往来有一定关系。To explore sequence characteristics of r NDA- ITS2 in Culex modestus at Alataw Pass, China- Kazakhstan and to analyze the genetic variation of Culex modestus for providing the reference for the Culex classification research and scientific prevention and control of mosquito- borne diseases. Genomic DNA was extracted from Culex modestus at different sites, and the r NDA- ITS2 gene of Culex modestus were amplified by PCR. The sequence analysis by BLAST and phylogenetic analysis by Mage 6.0. We obtained the r NDA- ITS2 sequences were approximate 323 bp by PCR. Blast showed that the Culex modestus was high homology with Culex restuans(U22136). There were transition,transversion and deletion in themono nucleotide of r NDA- ITS2 gene in the 24- 323 bp. r NDA- ITS2 gene sequence of Culex modestus were reported in this study. The r NDA- ITS2 gene of Culex modestus is genetic polymorphisms and they are probably associated with the ecological environment, geographical area and trade.
