摘 要:利用抑制性消减杂交(Suppression subtractive hybridization,SSH)方法构建了中国草原红牛与中国荷斯坦牛背最长肌差异表达消减c DNA文库,共获得859个差异表达克隆,对全文库进行了测序并与Gen Bank数据库进行比对分析。结果表明:共获得789个新ESTs,84个未知功能蛋白基因,356个已知功能蛋白基因。在已知功能蛋白基因中,表达上调的基因有123个,表达下调的基因有233个。差异表达基因可能与肉质性状有关,可以作为肉质性状的候选基因进行深入研究。Suppression subtractive hybridization (SSH) was used to construct cDNA library of the Chinese Red Steppes and Chinese Holstein Cattle dorsi muscle. Eight hundred and fifty-nine differentially expressed clones obtained from the library were sequenced and analyzed with blasting program of GenBank for sequence comparison. The results showed that a total of 789 new ESTs, 84 unknown genes and 356 functional genes were obtained in this library. Among these functional protein genes, there are 123 up-regulated genes and 233 down-regulated genes in Red Steppes cattle compared with Chinese Holstein cattle. These differentially expressed genes may be related to meat quality traits and can be used as candidate genes for meat quality traits.
