生精胶囊改善雄性去势大鼠勃起功能的研究

摘　　要：目的研究生精胶囊对雄性去势大鼠勃起功能的影响。方法成年雄性SD大鼠40只,按随机数字法随机分为假手术组、去势组、生精胶囊组和睾酮组。假手术组作为正常对照,其余组均行双侧睾丸切除术建立勃起功能障碍模型。术后假手术组、去势组给予蒸馏水灌胃,生精胶囊组给予310.2mg/kg·d^(-1)剂量的生精胶囊混悬液每天灌胃一次,睾酮组给予十一酸睾酮26.3mg/kg肌注,4周一次。每周记录各组大鼠体质量。干预8周后检测阴茎海绵体内压(intracavemosal pressure,ICP),测定血清睾酮水平以及称量前列腺和精囊腺组织。结果生精胶囊组与去势组间体质量、前列腺质量、精囊腺质量及血清睾酮水平比较,差异均无统计学意义(P均>0.05);而较假手术组显著降低,组间比较差异均有统计学意义(P均<0.01)。生精胶囊组ICP/平均动脉压(mean arterial pressure,MAP)为(0.55±0.073)mmHg,显著高于去势组(0.28±0.061)mmHg,差异有统计学意义(P<0.005),但显著低于假手术组(0.69±0.055),差异有统计学意义(P<0.05)。结论生精胶囊可以显著提高雄性去势大鼠ICP,改善勃起功能。Objective To evaluate the effects of the Chinese herbal formula Shengjing Capsule on castrated erectile dysfunction （ED） in a male rat model. Methods Forty adult male Sprague-Dawley rats were randomly divided into four groups, including sham-operated group, castrated group, Shengjing capsule group and testosterone group. Rats in sham-operated group served as the normal control, and rats in other groups were subjected to bilateral orchiectomy to establish the rat models of erectile dysfunction （ED）. One week after surgery, the rats in sham-operated group and castrated group were intragastrically administrated with distilled water, and rats in Shengjing capsule group and testosterone group were administrated with 310.2 mg/kg？d-1 shengjing capsule suspension by gavage and injected intramuscularly with 26.3 mg/[kg？month] testosterone undecanoate, respectively. The body weights of rats in each group were recorded weekly. At 8 week after treatment, the erectile function of the rats was assessed by measuring intracavernosal pressure （ICP） during electrostimulation of the cavernosal nerve. The prostate gland weight, seminal vesicles weight and serum testosterone levels were also evaluated. Results Changes in body weight, prostate gland weight, seminal vesicles weight and serum testosterone were not significant between the rats of Shengjing capsule group and castrated group （P=0.268, 1.00, 1.00, 1.00, respectively）. The Shengjing capsule group had the significant decrease in these parameters compared with the sham-operated group （P〈0.01 for all）. The ratio of the ICP to the MAP （mean arterial pressure） was significantly higher in the Shengjing capsule rats （0.55±0.073） than that in the models （0.28±0.061） （P=0.000）. But the ratio was significantly lower in the Shengjing capsule rats than that in the sham-operated group （0.69±0.055） （P=0.000）. Conclusion Oral treatment with Shengjing capsule for 8 weeks can increase the ICP and improve erectile function potentially.
