一锅法合成医药中间体异吲哚并1,2,3-三唑的研究

摘　　要：以Pd（PPh3）2Cl2/selectfluor为催化剂,在Et3N/THF/Me CN混合溶剂中,利用叠氮与炔烃的连串反应合成异吲哚并1,2,3-三唑医药中间体。研究了催化剂种类及其用量、反应温度、反应底物种类等因素对反应的影响,通过对产物进行GC-MS和NMR分析,得到了合成异吲哚并1,2,3-三唑的最佳反应条件为：邻碘苄基叠氮0.3 mmol,苯乙炔1.2 equiv,溶剂Et3N/THF/Me CN（1∶1∶1）3 mL,在Pd（PPh3）2Cl2（5mmol%）/selectfluor（2 equiv）的催化下,80℃条件下反应回流,产率达到58%-85%。Isoindoline fused with triazoles were selecffluor catalyst via cascade reaction of azides and synthesized in Et3N/THF/MeCN solvent with Pd （ PPh3 ）2Cl2/ alkynes. The effects of sorts and amounts of catalysts and reaction temperature on the yields were investigated, the influence of structure of substrate on tne reaction tolerance was researched as well. The structures of products were characterized by melting points and NMR. The results showed that the yield of isoindoline fused with triazoles was 58% - 85% under the optimum condition that 2-ido-benzyl azides 0. 3 mmol, alkynes 1.2 equiv, solvent Et3N/THF/MeCN （ 1 ： 1 ： 1 ） 3 mL, Pd （ PPh3 ） 2 C12 （ 5mmol% ）/selectfluor （ 2 equiv）, under refluxed at 80 ℃.
