摘  要:目的运用3D超声照射大鼠头部不同时间后,探讨其脑组织神经元细胞超微结构的变化。方法将12只大鼠随机分为六组,除对照组外,其他组均运用3D超声分别予以大鼠头部10、20、30、40min及50min的照射,3d后脱臼处死,取出大鼠脑部,用2%戊二醛固定24h,透射电镜常规制片,透射电镜下观察、拍照。结果 3D超声照射大鼠头部10、20min后,脑组织神经元细胞的超微结构未见明显变化。照射30min以后,胞浆内见线粒体、内质网、少量溶酶体,并见线粒体极排列紊乱、少量线粒体空泡变性。照射50min后,异染色质增多,胞浆内见线粒体、内质网,线粒体空泡变性、电子密度减低。结论 3D超声照射大鼠头部20min内,大鼠脑组织神经元细胞的超微结构未见明显变化,照射30min以后,大鼠脑组织神经元细胞超微结构受损,并随照射时间延长递进增加。Objective To investigate the changes of the ultrastructure of neurons in the brain of rats after different times of the head 3D ultrasound scanning.Methods 12 rats were randomly divided into six groups,the head of the rats were scanned 10,20,30,40 and 50min by using 3D ultrasound scanning except the control group,the rats were sacrificed by cervical dislocation after three days,and removed the brain,fixed with 2% glutaraldehyde for 24 h,prepared the slides by TEM(transmission electron microscope)conventional techniques,observed and photographed under TEM.Results There were no significant differences or changes in the ultrastructure of brain neurons after 10 or 20minutes 3D ultrasound exposed.The mitochondria,endoplasmic reticulum and a small amount of the lysosomal were found in cytoplasm,there was a disarranged mitochondrial cristae and a small amount of vacuole degeneration after 30 minutes 3D ultrasound exposed.There were heterochromatin increased,intracytoplasmic mitochondria,endoplasmic reticulum,vacuolar degeneration of mitochondria,electron density reduced after 50 minutes 3D ultrasound exposed.Conclusions Rat brain ultrastructure of neurons show no obvious change within 20 minutes 3Dultrasound exposed,damage and progressive increase extent of damage with prolonged exposed time after 30 minutes.
