摘  要:目的评价迷走神经刺激对老龄大鼠术后认知功能障碍的影响。方法清洁级健康SD大鼠30只,18~20月龄,体重390~550g,采用随机数字表法,将其分为3组(/2=10):对照组(c组)、手术组(s组)、迷走神经电刺激组(V组)。S组和V组行右颈动脉鞘切开暴露迷走神经术,V组行迷走神经电刺激(波宽1ms、频率10Hz、电压1~2V、持续时间30min)。术前4、3、2、1d、术后2d进行水迷宫定位航行实验。术后2d进行空间探索实验和日广场实验测定认知功能,记录逃避潜伏期、穿越平台次数、跨格次数、直立次数和中央格停留时间。行为学实验结束后,经颈静脉采集血样,采用ELISA法检测血清TNF-α、IL-1β和IL-6的浓度。结果与C组比较,S组术后2d时穿越平台次数、跨格次数及直立次数减少,逃避潜伏期和中央格停留时间延长,血清TNF—α、IL-1β和IL-6的浓度均升高(P〈0.05);与s组比较,V组大鼠术后2d时穿越平台次数、跨格次数及直立次数增加,逃避潜伏期和中央格停留时间缩短,血清TNF-α、IL-1β和IL-6的浓度均降低(P〈0.05)。结论颁动脉鞘迷走神经电刺激可改善老龄大鼠术后认知功能障碍。Objective To evaluate the effect of electric vagal stimulation on postoperative cognitive dysfunction in aged rats. Methods Thirty healthy Spraguc-Dawley rats of botb sexes, aged 18- 20 months, weighing 390-550 g, were randomly divided into 3 groups (n= 10 each) using a random number table: control group (group C), surgery group (group S), and electric vagal stimulation group (group V). Incision of the right carotid sheath was perfbrmed, and the vagal nerve was exposed after anesthesia in S and V groups. The right vagus nerve was stimulated for 30 rain with continuous electric rectangular pulses (1ms, 10 Hz, 1-2 V) after surgery in group V. The Morris water maze place navigation test was performed at 4, 3, 2 and 1 days betore surgery and 2 days after surgery. The cognitive tunctiou was assessed using spatial probe and open field tests conducted at 2 days after surgery. The escape latene, y, the plumber of crossing the platform, the number of crossing the grid, the number of standing on the back legs, and the time animals spent in the central square were recorded. After the end of behavioral testing, venous blood samples were collected from the jugular vein for determination of concentrations of serum tumor necrosis factor-alpha (TNF-α) , interleukin (IL) -1β and IL-6 using enzyme-linked immunosorbent assay. Results Cmnpared with group C, the number of crossing the platform, the number of crossing the grid, and the number of standing on the back legs were significantly decreased, the escape latency and the time animals spent in the central square were significantly prolonged, and the concentrations of serum TNF-α, IL-1β and IL-6 were significantly increased at 2 days after surgery in group S (P〈0.05). Compared with group S,the number of crossing the platform, the number of crossing the grid, and the number of standing on the back legs were significantly increased, the escape latency and the time animals spent in the central square were significantly shortened, and the concen
