摘  要:目的为了探究具有免疫抑制和抗肿瘤作用的甲氨蝶呤(MTX)对干扰素(IFN)的影响,以及IFN在风湿免疫性疾病中的作用。方法以5、20、100μg/mL浓度梯度的MTX作用于Huh7.5.1细胞48h,采用实时荧光定量PCR法定量检测MTX干预后Huh7.5.1细胞内IFN-α、IFN-β、IFN-γ、干扰素刺激基因15(ISG15)、抗黏病毒蛋白(MxA)的mRNA表达量。结果 Huh7.5.1细胞在MTX(5、20μg/mL)干预下内源性IFN-α、IFN-β的mRNA表达量与对照组相比均明显上升,差异均有统计学意义(P<0.05),并且随着MTX干预浓度的上升而上升;MTX浓度为100μg/mL时,与对照组相比虽有上升但差异无统计学意义(P>0.05)。而IFN-γ的mRNA表达量仅在MTX浓度为20μg/mL时与对照组比较差异有统计学意义(P<0.05),在5、100μg/mL时虽有上升但差异无统计学意义(P>0.05)。在MTX干预后,MxA和ISG15的mRNA表达量均有不同程度的上升(P<0.05)。结论小剂量MTX治疗风湿免疫性疾病取得良好的疗效与IFN相关。Objective To explore the effect of methotrexate(MTX),which has immunosuppressive and antitumor properties On interferon production and the role of interferon(IFN) in rheumatic autoimmune disease. Methods Cutured HuhT. 5.1 cells were treated with different concentration of MTX(5,20,100 μg/mL) for 48 hours. After MTX intervention, the mRNA expression levels of IFN-α, IFN-β, IFN-γ, ISG15, MxA in HuhT. 5.1 cells were quantified by real-time quantitative polymerase chain reaction. Results Compared with the control group,the endogenous IFN-α,IFN-β mRNA expressions of Huh7.5.1 cells under the intervention of methotrexate(5, 20 μg/mL) were significantly increased,the differences were statistically significant(P〈0. 05) ,which was inereaing with the rise of the concentration of MTX intervention. MTX concentration of 100 μg/mL, compared with the control group, although there was an increase, but without statistical significance(P〉0.05). While the mRNA expression of IFN-α was only significantly different when the MTX concentration was 20 μg/mL(P〈0.05). Although there was an increase in 5 μg/mL and 100μg/ mL, there was no statistical significance(P〉0.05). In MTX, the expression levels of MxA and ISGI5 were increased in different degrees(P〈0.05). Conclusion The good efficacy of low-dose MTX in the treatment of rheumatic autoimm.une diseases maybe related to IFN.
