针刀干预对大鼠膝内侧副韧带瘢痕组织中胶原含量及形态学的影响

摘  要:目的:探讨针刀对大鼠膝内侧副韧带(MCL)瘢痕组织模型中胶原含量及形态学的影响。方法:将15只健康SD雄性大鼠随机取3只为正常组,另12只造模,手术横断双侧膝内侧副韧带,造模第4周后,随机取3只处死,取韧带瘢痕组织做组织学观察,示造模成功,剩余9只按随机分为模型组、针刀组、激素组,针刀组、激素组分别用针刀、激素干预,1周后处死,观察HE、天狼星红、Smad3染色结果。结果:针刀组较模型组炎性细胞、微血管减少,水肿减轻,组织更致密;针刀组较模型组总胶原含量和Smad3均降低(P<0.01),针刀组与模型组Ⅰ、Ⅲ型胶原含量均无显著性差异(均P>0.05)。结论:针刀治疗可以减少大鼠膝内侧副韧带瘢痕组织模型中的总胶原含量,改善胶原纤维的排列顺序,其作用机制可能是通过减少Smad3的表达而实现的。Objective:To investigate the effect of acupotomy on the collagen content and morphology characteristics of medial collateral ligament scar in rats.Methods:Fifteen healthy male SD rats were randomly assigned to normal group and modeling group with a ratio 1:4.Rats in modeling group were surgically transected of bilateral knee medial collateral ligaments.After 4 weeks of surgery,the ligament scar tissues were taken for histological investigation from 3 rats to confirm the success of modeling.Then nine rats were randomly divided into the control modeling group,the acuputomy group,and the glucocorticoid hormone treatment group.One week after acuputomy or steroid hormone intervention,the ligament tissues were harvested and investigated by HE staining,Sirius red and Smad3 staining.Results:Compared with the model control group,less inflammatory cells,capillary,and edema were found in the acuputomy group.A lower level of the total collagen content and the expression of Smad3 was found in acuputomy group(P 〈0.01).Conclusion:The acupotomy treatment can decrease the total collagen content of rat knee medial collateral ligament scar tissue and improve the order of the arrangement of collagen fibers.
