摘 要:目的筛选糖尿病心肌病患者血清差异表达的micro RNA(mi RNA),明确mi R-186-5p和mi R-516a-5p在DCM发生发展中的作用和机制。方法选择30例DCM患者为实验组,60例年龄相当的身体健康者为正常对照组,分别从两组随机选取3例作为研究对象行micro RNA芯片初筛差异表达mi RNA,然后荧光定量q PCR验证这些mi RNA的差异表达;在高糖诱导人AC16心肌细胞损伤模型中,mi R-186-5p和mi R-516-5p的mimics和inhibitors分别干预,观察对高糖引起的心肌细胞毒性的影响。结果 micro RNA芯片检测发现,糖尿病心肌病患者血清有51个差异表达的mi RNA,经荧光定量q PCR验证为mi R-186-5p、mi R-516a-5p等8个差异表达mi RNA;显著高表达的mi R-516-5p和明显低表达的mi R-186-5p介导了高糖所致心肌细胞毒性损伤。结论糖尿病心肌病患者血清mi RNA表达谱发生改变,有8个差异表达mi RNA,mi R-516a-5p和mi R-186-5p可能参与糖尿病心肌病的心肌损伤发病过程。Objective To screen out serum differentially expression microRNAs and clarify potential roles of miR-186-5p and miR-516a-5p in diabetic cardiomyopathy patients. Methods 30 DCM patients were included into experimental group,60 healthy adults were recruited into control group. To screen serum differentially expression microRNAs in 3 DCM patients by microRNA microassay analysis and then verify using qPCR.To explore the effects of miR-186-5p or miR-516-5p mimics or inhibitors on cardiotoxicity induced by high glucose in human AC16 cells. Results 51 differentially expression microRNAs in serum of DCM patients were found by miRNA chip test, 8 dysregulated microRNAs were verified using q PCR. Upregulated miR-516-5p and downregulated miR-186-5p mediated the cardiotoxicity induced by high glucose in human AC16 cells. Conclusion There are 8 differentially expression microRNAs in DCM patients serum.miR-516a-5p and miR-186-5p mediated the cardiotoxicity induced in DCM.
