摘 要:以湖北百合(Lilium henryi Baker)鳞片为材料,将鳞片接种于4种不同配方的芽诱导培养基中,筛选出最佳配方,用此配方诱导出丛生芽,以丛生芽为诱变材料,在无菌条件下,将浸透秋水仙素溶液浓度分别为0.05%、0.10%、0.15%、0.20%的脱脂棉覆盖在新芽上,分别处理24、36、48 h。结果表明,芽诱导的最佳配方为6-BA 1.0 mg/L+NAA 0.5 mg/L,0.10%的秋水仙素处理48 h的诱变效果最佳,诱变率达20%。植株外观形态及气孔鉴定结果显示,多倍体植株叶片厚且增大,叶色加深,气孔的长、宽明显变大,保卫细胞密度减小。根尖染色体鉴定显示,四倍体植株的染色体数为48条(2n=4x=48),而二倍体的植株染色体数为24条。The scales of Lilium henryi Baker were inoculated in four bud induction medium with different formula to screen the best medium which was used to induce multiple shoots so as to provide material for mutagenesis. The buds were covered by absorbent cotton saturated with colchicine solution at different concentration(0.05 %,0.10 %,0.15 %,0.20 %) for 24,36,48 h under sterile conditions. The results showed that the best bud induction formula was 6-BA 1.0 mg / L+NAA 0.5 mg / L;while the most effective mutagenic conditions were treating with 0.10 % colchicine solution for 48 h,of which the mutation rate was 20 %.According to the identification result of plant morphology and stomata, leaf thickness of polyploidy plants increased; and the color was darken; length and width of stomata was significantly larger; density of guard cells decreased. According to chromosome observation of root tip, the chromosome number of tetraploid plants were 48(2n=4x=48);of diploid plants were 24.
