摘 要:本试验旨在研究L-精氨酸(Arg)和α-酮戊二酸(AKG)对热应激肉鸡肠道功能的影响。选用200只1日龄Cobb肉鸡,随机分为4个处理组:对照组(基础日粮)、热应激组(基础日粮)、Argg(基础日粮+0.5%Arg)、AKG组(基础日粮+0.5%AKG),每个组5个重复,每个重复10只鸡。试验为期5周,第1周各组室温控制在32-34℃,第2~5周热应激组、Arg组和AKG组控制在(36±2)℃,而对照组第2周开始慢慢降至(22±2)℃,并从第3周开始维持在(22±2)℃,湿度为50%-60%。结果表明:与对照组相比,热应激处理降低了肉鸡生长性能和血液三碘甲腺原氨酸(T1)含量(P〈O.05),同时也显著降低了空肠和回肠脂肪酶、回肠胰蛋白酶、空肠过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH—Px)、回肠超氧化物歧化酶(SOD)和CAT的活力、十二指肠和回肠三磷酸腺苷(ATP)含量、空肠小肽转运载体(PepTl)、紧密连接蛋白(Claudin-5)mRNA表达量(P〈0.05),显著提高了死亡率、血液皮质酮、十二指肠和回肠丙二醛(MDA)含量、空肠低氧诱导因子-1α(HIF-1α)、AMP依赖的蛋白激酶d1(AMPK-α1)mRNA表达量(P〈O.05);与热应激组相比,日粮中添加0.5%Arg可显著提高热应激肉鸡饲料转化率、十二指肠脂肪酶和回肠胰蛋白酶活力、十二指肠和空肠DNA含量、空肠CAT活力和HMOX—lmRNA表达量(P〈0.05),降低死亡率、十二指肠和空肠RNA/DNA比值、空肠TP/DNA比值、HIF-1α和AMPK—α1 mRNA表达量(P〈0.05),而日粮中添加0.5%AKG可显著提高热应激肉鸡血液L、十二指肠和空肠DNA含量、十二指肠CAT和GSH—Px的活力、十二指肠和空肠及回肠ATP含量(P〈0.05),并显著降低死亡率、血液皮质酮含量、空肠RNA/DNA及TP/DNA比值、十二指肠MDA和回肠过氧化氢(H202)含量(P〈0.05)。因�This study was conducted to investigate the effects of arginine and α-ketoglutaric acid on intestinal function of heat-stressed broilers. 200 1-day-old Cobb chickens were randomly divided into four groups (control group, heat- stress group, Arg group, and AKG group). Each treatment had 5 replicate pens with 10 chickens per pen. The control group and heat-stress group were fed a basal diet, whereas Arggroup were fed basal diet + 0.5% Arg and AKG group were fed basal diet +0.5% AKG. The heat-stressed process was followed as: first week 32-34 ℃, second to fifth week (22 ± 2) ℃ (the control group) and (36±2) ℃ ( other groups). The humidity of all groups was 50%-60%. The results showed that: Compared with the control group, heat-stress treatment decreased the growth performance, the death rate, and the level of T3 in blood, as well as the activities of lipase in the jejunum and ileum, the trypsin activity in the ileum, the activities of CAT and GSH-Px in the jejunum, the activities of SOD and CAT in the ileum, the levels of ATP in the duodenum and ileum and the Claudin-5 and PepT1 mRNA levels in the jejunum, but increased the level of corticosterone in blood, the levels of MDA in the duodenum and ileum, the HIF-1α and AMPK-α mRNA levels in the jejunum. Compared with heat stress group, diet supplemented with 0.5% Arg increased the lipase activity in the duodenum, the trypsin activity in the ileum, the levels of DNA in the duodenum and jejunum, the CAT activity in the jejunum, and the HMOX-1 mRNA level in jejunum, but decreased the death rate, the ratios of RNA/DNA in the duodenum and jejunum, the ratio of TP/DNA in the jejunum, the HIF-1α and AMPK-cd mRNA level in the jejunum of heat-stress broilers.
