摘 要:目的建立同时测定阔叶十大功劳叶药材中盐酸小檗碱、盐酸巴马汀、盐酸药根碱的高效液相色谱(HPLC)法,并对不同产地阔叶十大功劳叶药材进行测定与比较。方法色谱柱采用Welch Ultimate LP—C18柱(150mm×4.6mm,5μm),流动相为0.05mol/L磷酸二氢钾(用磷酸调节pH至3.0)-乙腈(73:27),流速1.0mL/min,检测波长346nm,柱温30℃。结果盐酸小檗碱、盐酸巴马汀、盐酸药根碱色谱峰达到基线分离,质量浓度分别在5.328—266.4μg/mL(r=0.9999),3.68—184.0μg/mL(r=0.9999),2.412~120.6μg/mL(r=0.9998)范围内与峰面积呈良好的线性关系,平均加样回收率分别99.17%(RSD=0.35%,n=6),98.97%(RSD=0.65%,n=6),99.36%(RSD=0.44%,n=6)。结论该法操作简单、准确可行、重复性好,可用于阔叶十大功劳叶药材的质量控制。Objective To establish an HPLC method for simultaneous determination of 3 alkaloids (berberine, palmatine, jatrorrhizine) in Mahonia bealei(Fort. ) leaves from different regions. Methods The analysis was carried out on a Welch Ultimate Lp-C18 column with a mobile phase consisted of acetonitrile and 0.05 mol/L monopotassium phosphate solution (add phosphoric acid to pH =3.0)(27:73). The column temperature was 30℃ and the detection wavelength was set at 346 nm and the flow rate was 1.0 mL/min. Results There was a good linear relationship in the range of 5.328-266.4 μg/mL(r=0.999 9), 3.68-184.0 μg/mL(r=0.999 9), 2.412 - 120. 6 μg/mL( r = 0. 999 8), respectively, and the average recovery rates ware 99. 17% ( RSD = 0.35%, n = 6), 98.97% ( RSD = 0. 65%, n =6), 99.36% (RSD =0. 44%, n =6), respectively. Conclusion The method is simple and well reproducible, and can be readily utilized as a quality control method for Mahonia bealei Leaves.
