摘 要:目的考察以大豆磷脂及胆固醇作为膜材料,用薄膜-水化法制备脂质微泡的最佳工艺条件,并通过对脂质微泡粒径分布的考察筛选最佳处方工艺。方法以大豆磷脂、胆固醇为成膜材料,六氟化硫(SF6)气体为核心物质,采用薄膜-水化法制备脂质微泡;以D_(0.9)为指标,D_(0.9)越小,说明符合要求的微泡越多;采用控制变量法考察不同投料比、超声功率与时间、PEG分子量对微泡粒径分布的影响。结果通过对粒径分布的测定及比较,确定了制备六氟化硫脂质微泡的最佳工艺,并考察了在该工艺条件下制备出微泡的其他理化性质。结论所制备的六氟化硫脂质微泡的形态规则、大小均匀、Zeta电位、包封气体量及半衰期均符合要求。OBJECTIVE To study the optimum conditions for the formation of microbubble by using soybean phospholipid and cholesterol as film forming material. METHODS Using sulfur hexafluoride as the core material, lecithin and cholesterol as film materials liquid microbubble was prepared by thin - film rehydration method. D0.9 value represented 90% of microbubble within optimized size, so the D0.9 value should be as small as possible. The influence on microbubbles' diameter distribution by changing different ratios of lecithin and cholesterol, ultrasonic power and time, and molecular weight of PEG were investigated by the controlling variables method. RESULTS By measuring and comparing distribution of microbubbles' diameter, the optimal process for preparing sulfer hexafluoride liquid microbubble was determined and other physical and chemical properties of microbubble prepared in the same process were examined. CONCLUSION Microbubble prepared by this method has reqular morphology and uniform size, and its Zeta potential, encapsulation efficiency and half- life all meet requirements by this process.
