摘  要:以木榄幼叶为外植体,进行其愈伤组织诱导的影响因子研究,通过灭菌条件、培养基成分、添加剂种类、外源植物激素浓度试验,比较外植体褐化率与愈伤组织诱导率。结果表明:75%乙醇灭菌30s后在2%(体积分数v/v)NaClO溶液灭菌20min是木榄幼叶表面消毒适宜条件;氨基酸培养基是基础培养基;45℃热激与添加1.0mmol·L^(-1)抗坏血酸均可降低褐化率;AA培养基+0.50mg·L^(-1) 2,4-D+0.50mg·L^(-1) TDZ+1.00mmol·L~(^(-1))维生素C或AA培养基+0.50mg·L^(-1) 2,4-D+0.50mg·L^(-1) CPPU+1.00mmol·L^(-1)维生素C均可得大于50%的愈伤诱导率。因此,克服外植体褐化与应用CPPU或TDZ作为细胞分裂素是木榄愈伤组织诱导成功的关键。The factors affecting callus initiation from magrove plant Bruguiera gymnorrhiza was investigated with the young leaves as explant.Sterilization conditions,medium components,kinds of additive,different concentration of plant growth regulator combination were tested and callus induction rate was calculated.The results showed that 75%alcohol30seconds with 2%(volumn fraction w/v)NaClO 20 minutes was the best condition of surface sterilization;amino acid(AA)medium was basic medium;browning rate was inhibited by heat-shock at 45 ℃ and ascorbic acid;AA medium+0.50mg·L^-1 2,4-D+0.50mg·L^-1 TDZ+1.00mmol·L^(-1) vitamin C or AA medium+0.50mg·L^-1 2,4-D+0.50mg·L^-1 CPPU combination were optimal callus induction medium,in which over 50%of callus induction rate was obtained.Therefore,explant browning prevention and application of CPPU or TDZ as cytokinine were the key to success in callus induction fromBruguiera gymnorrhiza.
