摘 要:目的:探讨食管鳞状细胞癌放射抗拒细胞株特征性蛋白指纹图谱,为进一步的临床研究提供依据。方法:确定食管鳞状细胞癌细胞株(ECa109)的亚致死剂量射线。构建食管鳞状细胞癌放射抗拒细胞株模型(ECa109-IR)。克隆生存实验、细胞增殖实验和流式细胞术检测ECa109-IR具有放射抗拒性。将食管鳞癌放射抗拒细胞株(ECa109-IR)与食管鳞癌放射敏感细胞株(ECa109)送至北京进行蛋白指纹图谱的检测。结果:成功构建食管鳞状细胞癌放射抗拒细胞株(ECa109-IR)。克隆生存实验、细胞增殖实验、流式细胞术证实ECa109-IR具有放射抗拒性。蛋白指纹图谱表明:ECa109-IR细胞和ECa109细胞的蛋白质荷比峰存在明显差异(P<0.05的差异峰为21个,P<0.01的差异峰为19个),21个差异表达的蛋白质峰中,13个质荷比峰在ECa109-IR中表达下调,8个质荷比峰在ECa109-IR中表达上调。结论:ECa109-IR和ECa109蛋白指纹图谱之间有21个存在明显差异的蛋白峰(即两者之间存在差异蛋白),这些差异蛋白峰可能与肿瘤细胞放射抗拒相关,可为食管癌的临床研究提供依据。本研究的不足之处为未进行差异表达蛋白的分离鉴定,下一步的计划是鉴定具体蛋白,提出食管鳞癌放疗敏感性的预测模型。Objective:To study the proteomic fingerprint characteristic of ECa109-IR.Methods:Confirm the sublethal ionizing radiation dose of ECa109.To establish a radioresistant subclone cell line-ECa109-IR.Using the clonal survival test,cell proliferation assay and flow cytometry to test the radioresistant of ECa109-IR.For radioresistance ECa109-IR and radiosensitivity ECa109 to detect the protein spectrum using protein fingerprint technique.Results:Established a radioresistant subclone cell line-ECa109-IR successfully.The clonal survival test,cell proliferation assay and the flow cytometry showed that the ECa109-IR has radiation resistance.The protein fingerprint of ECa109-IR and ECa109 showed that:There were obvious differences in ECa109-IR and ECa109 protein M / Z peak( the peak was 21 when P〈0.05,and the peak was 19 when P〈0.01).And 13 M/Z peaks were down regulated and8 M / Z peaks were up regulated.Conclusion:There were 21 obvious different protein M / Z peaks between ECa109-IR and ECa109,in other words,there were different protein between them.The different protein M / Z peaks may be connect with the radioresistance and can provide basis for clinical study.But the study also has defect,we didn't isolate and identify the protein M / Z peaks.And the next is to identify concrete protein and structure the prediction model of esophageal cancer radiosensitivity.
