摘  要:目的探讨甲强龙(MP)对大鼠脊髓损伤(SCI)后神经细胞凋亡的影响,以及相关分子机制研究。方法 45只SD大鼠随机分为3组,对照组(n=15),模型组(n=15)和实验组(n=15)。实验组于术后给与甲强龙尾静脉注射(30 mg/kg),模型组为SCI模型,对照组只切开椎板,不损伤脊髓。分别于7 d、14 d和21 d进行下肢功能评分(BBB评分);21 d后取出损伤脊髓,利用Western blot法检测Bax和Bcl-2蛋白表达;免疫组织化学染色及TUNEL染色检测Caspase-3阳性细胞数和神经细胞凋亡水平。结果各时间点BBB评分,模型组显著低于对照组,而实验组明显高于模型组(P<0.05);Western blot法显示,甲强龙显著抑制Bax,提高Bcl-2表达(P<0.05);组织学观察显示,甲强龙显著抑制Caspase-3表达,以及下调TUNEL阳性细胞数(P<0.05)。结论甲强龙可通过抑制大鼠脊髓损伤Bax和Caspase-3表达,提高Bcl-2表达,从而抑制神经细胞的凋亡。Objectiye To explore the effect of methylprednisolone on spinal cord injury - induced neural cell apoptosis in rats and its po-tential mechanism. Methods 45 SD rats were divided into 3 groups(n = 15),the control group,the model group and the test group. The rats in test group was given by tail intravenous injection(30 mg / kg)after SCI,and rats only opened laminectomy without damaging the spinal cord in sham operation group. At the 21 d after operation,motor function of the rat hind limb was determined by the BBB(Basso,Beattie,Bresnahan, BBB);The protein expression of Bax and Bcl - 2 were detected by western blot at 21 d via western blot;The expression of caspase - 3 and the lev-el of neuronal cell were detected by immunohistochemistry and TUNEL methods. Results At different time of points,the BBB value in model group was markedly lower than that in the control group,while BBB value in the test group was significantly higher than that the model group( P〈 0. 05). Western blot showed that methylprednisolone could significantly suppress the protein expression of Bax and improve the Bcl - 2 expres-sion( P 〈 0. 05). Histological observation showed that methylprednisolone could markedly suppress the caspase - 3 expression and down - regu-late the cell apoptosis level( P 〈 0. 05). Conclusion Methylprednisolone can inhibit the expressi of Bax and Caspase - 3 of spinal cord injury in rats,and increase the expression of Bcl - 2,thereby inhibiting neuronal apoptosis.
