摘 要:目的研究苏子油对兔颈动脉粥样硬化干预作用及微小RNA21(miR-21)、基质金属蛋白酶9(MMP-9)基因表达的影响。方法用数字法将50只健康成年新西兰雄性大耳白兔随机分为正常组、模型组、对照组、高剂量及低剂量实验组,各10只。经150 g·d^(-1)高脂饲料喂养和颈动脉球囊损伤术处理建立颈动脉粥样硬化模型。对照组,给予阿托伐他汀钙2.5 mg·(kg·d)^(-1);低高2个剂量实验组,给予苏子油软胶囊0.15,0.60 g·(kg·d)^(-1);正常组、模型组的动物给予等量0.9%Na Cl灌胃。6周后,用病理图像分析系统观察右颈动脉血管壁(中膜)面积/血管腔面积之比(W/L),用胆固醇氧化酶法检测血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平,以磷酸甘油氧化酶法检测血清三酰甘油(TG)水平,用实时荧光定量PCR法检测miR-21、MMP-9基因表达水平。结果治疗后,正常组、模型组、对照组和高、低2个剂量实验组的右颈动脉血管W/L分别为(1.27±0.15),(6.42±0.81),(2.94±0.42),(2.88±0.41),(4.17±0.54),与模型组比较,各组动物上述指标差异有统计学意义(P<0.05);与低剂量组比较,高剂量组和对照组动物右颈动脉血管W/L差异有统计学意义(P<0.05),且高剂量组和对照组动物右颈动脉血管W/L比较差异无统计学意义(P>0.05)。治疗后,正常组、模型组、对照组和高、低2个剂量实验组的血清TC水平分别为(1.59±0.22),(22.45±3.17),(8.43±1.06),(8.39±1.04),(13.54±1.27)mmol·L^(-1),TG水平分别为(0.79±0.11),(2.86±0.38),(1.24±0.15),(1.21±0.12),(1.87±0.26)mmol·L^(-1),LDLC水平分别为(0.71±0.08),(15.38±2.14),(6.56±0.93),(6.51±0.89),(9.89±1.32)mmol·L^(-1),与模型组比较,各组动物上述指标水平差异有统计学意义(均P<0.05);与低剂量组比较,高剂量组和对照组动物血清TC、TG及LDL-C水平差异有统计学意义(均P<0.05),且高剂量组和对照组动物血清TC、TG及LDL-C水平比较差异无统计学意义(P>0.05)。治�Objective To investigate the effect of Perilla oil on the expressions of micro RNA21 (miR -21 ) and matrix metalloproteinases 9 (MMP -9 )genes in rabbit with carotid athero- sclerosis. Methods By digital method, fifty healthy adult male New Zealand white rabbits were randomly divided into normal group, model group, control group, high dose group and low dose test groups, 10 in each group. By feeding 150 g · d^-1 high fat diet and carotid artery balloon angioplasty, the carotid atherosclerosis model was established. Animals in control group were given atorvastatin calcium 2. 5 mg · (kg · d)^ -1, animals in low dose test group were given Perilla oil soft capsule 0. 15 g ~ ( kg ~ d) -1, animals in high dose test group were given Perilla oil soft capsule 0. 60 g · (kg · d)^ -1 by intragastric administration, and fed with high fat diet 150 g · d^-1 ;animals in normal group and model group were given equal volume of normal saline by intragastric administration, and fed with normal diet 150 g · d^-1. After 6 weeks treatment, the ratio of right carotid artery vascular wall ( medium film) area/vascular lumen (W/L) area was observed by pathological image analysis system. The cholesterol oxidase method was used to detect the serum total cholesterol (TC), low density lipoprotein cholesterol (LDL- C ) levels. And serum triglyceride (TG) levels were detected by the method of glycerol phosphate oxidase. The expression levels of miR -21 and MMP -9 genes were detected by real time fluorescence quantitative PCR method. Results After treatment, the W/L of right carotid artery in normal group, model group, control group, high dose and low dose test groups were (1.27 ±0. 15), (6. 42 ± 0. 81 ), (2. 94 ± 0. 42), (2. 88 ± 0. 41 ), (4. 17 ± 0. 54), compared with model group, the above indicators levels were lower in each group.
