摘 要:目的:通过建立动物模型,探讨姜黄素(CUR)是否可抑制环磷酰胺(CP)所致雄性昆明小鼠生殖毒性。方法:采用随机对照实验,将40只7~8周龄30~35g雄性昆明小鼠随机分为4组(每组10只):正常对照组(生理盐水),溶剂对照组(玉米油+10%二甲基亚砜),模型组(CP 50mg/kg),实验组(CUR 50 mg/kg+CP 50 mg/kg)。连续腹腔注射5d,最后一次注射后24h内顺次处死小鼠。测量小鼠体重及睾丸、附睾重量,计算睾丸、附睾脏器指数;获取血清检测睾酮水平;收集右侧附睾精细胞悬液进行精液分析。结果:正常对照组与溶剂对照组各指标比较差异均无统计学意义(均P〉0.05)。与正常对照组、溶剂对照组相比,模型组、实验组小鼠体重、睾丸重量、附睾重量减轻(均P〈0.05),精子计数减少(P〈0.05),血清睾酮水平降低(P〈0.05),死亡精子百分比、畸形精子百分比明显增加(P〈0.05),但实验组上述指标较模型组均有所改善(P〈0.05)。模型组睾丸脏器指数较正常对照组、溶剂对照组明显降低(P〈0.05),但附睾脏器指数比较差异无统计学意义(P〉0.05)。实验组睾丸脏器指数、附睾脏器指数与各组比较差异均无统计学意义(均P〉0.05)。结论:CUR可改善CP所致雄性昆明小鼠急性期生殖损伤。Objective: To explore the protective effect of curcumin (CUR) on reproductive toxicity inducedby cyclophosphamide (CP) in male mice. Methods: 40 male mice aged 7-8 weeks (30-35 g) were randomlydivided into four groups ( n = 1 0 per group) : (1) The normal control group in which the mice were givennormal saline; (2) the solvent control group in which the mice were given vehicle (10% DMSO in corno il); (3) the model group in which the mice were given 50 m g/kg CP; (4) the experimental group in whichthe mice were treated with 50 m g/kg CUR plus 50 m g/kg CP. All mice were administered for 5 consecutivedays via intraperitoneal injection, and were killed within 24 h after the last injection. The weight of thebody, testis and epididymis were measured, and the indexes of testis and epididymis were calculated. Thelevel of testosterone in the serum was determined. The sperm suspension of the right epididymis was collectedto conduct semen analysis. Results: There were no significant difference in all indexes between thenormal control group and the solvent control group ( P 〉0. 05). Compared with the normal control groupand the solvent control group, the weight of the body, testis and epididymis, as well as the sperm countand serum testosterone level were decreased in the model group and the experimental group ( P 〈0. 05),while the percentage of dead and abnormal sperm was increased. The above indexes were significantly improvedin the experimental group when compared with the model group( P 〈0. 05). The index of testis in the model group was lower than that in the normal control group and the solvent control group, but there was no significant difference in epididymis index among groups ( P 〉 0 . 05). There was no difference in both indexes of testis and epididymis between the experimental group and any other groups ( P 〉 0 . 05). Conclusion: CUR could ameliorate CP-induced reproductive toxicity in male mice.
