摘 要:目的:观察二甲双胍对哮喘模型小鼠气道炎症的影响。方法:将24只健康BALB/c雌性小鼠,按随机数字表法分为正常对照组、二甲双胍干预组及哮喘组,每组各8只。二甲双胍干预组及哮喘组于实验第0天、第14天经腹腔注射50μg卵蛋白(OVA)及2mg氢氧化铝致敏,共2次,并于实验第21天开始吸入5%OVA雾化激发,连续14d,二甲双胍干预组于雾化前30min按5mg/只小鼠剂量经腹腔注射二甲双胍,每周3次。实验第35天处死小鼠,收集支气管肺泡灌洗液(BALF)并检测其有核细胞总数及分类计数;分离肺组织行HE染色,光镜下观察肺组织病理变化;采用流式细胞术检测脾脏Th17细胞的表达;采用ELISA法检测小鼠BALF中细胞因子IL-17的水平。结果:二甲双胍干预组BALF有核细胞总数、嗜酸性粒细胞百分比(EOS%)、中性粒细胞百分比(NEU%)均显著低于哮喘组(P<0.05);二甲双胍干预组小鼠肺组织病理变化较哮喘组明显减轻;二甲双胍干预组脾脏Th17细胞占CD4+T细胞的比例低于哮喘组(P<0.05);二甲双胍干预组BALF中IL-17水平低于哮喘组(P<0.05)。结论:二甲双胍可以通过抑制Th17细胞表达,从而减轻哮喘小鼠的气道炎症。Objective To investigate the effect of metformin on the treatment of airway inflammation inasthma mice. Methods: 24 female BALB/c mice were divided into the normal control group, the metformingroup and the asthma group according to a random number table ( n = 8 per group). The mice in the m etformingroup and the asthma group were given 50μg ovalbumin (OVA ) and 2 mg aluminium hydroxidei. p. on day 1 and day 14, and then were exposed to 5% OVA aerosol from day 21 to day 34. The mice inthe metformin group were treated with metformin at 30 min before atomization (i. p . , three times aw eek). The mice were sacrificed on day 35 , and the bronchoalveolar lavage fluid (BALF) was collected andin which the nucleated cell counts were recorded. The pathological changes of lung tissue were observed by HE staining. The expression of T h17 in spleen was measured by flow cytom etry, and the level of IL-17 inBALF was determined by ELISA. Results: The amount of nucleated cells, percentage of eosinophil andneutrophil in BALF in the metformin group were lower than those in the asthma group ( P 〈0. 05). Lungpathologic changes were improved in metformin group compared with that of the asthma group. IL-17 levelin BALF in the metformin group was lower than that, of the asthma group ( P 〈0. 05). The expression of Th17 in spleen in the metformin group was lower than that of the asthma group ( P 〈0. 05). Conclusion:Metformin can attenuate airway inflammation in asthma mice through inhibiting the generation of Th17 cells.
