摘 要:目的:研究凌霄花总黄酮对脑缺血再灌注损伤大鼠的保护作用,并探讨其作用机制。方法:SPF级Wistar大鼠60只,随机分为假手术组,模型组,阳性药组(阿司匹林片,50 mg·kg^(-1)),凌霄花总黄酮高、低剂量组(200,100 mg·kg^(-1)),除假手术组外,建立短暂性大脑中动脉阻塞大鼠模型(t MCAO),采用神经缺损评分(NDS)法评价行为学的改变、苏木素-伊红(HE)染色法评价脑组织病理形态的改变、酶联免疫吸附测定(ELISA)检测血清中神经元特异性烯醇化酶(NSE)的水平及脑组织中细胞因子白细胞介素-10(IL-10),转化生长因子-β_1(TGF-β_1)的水平,探讨凌霄花总黄酮对脑缺血再灌注损伤的保护作用。结果:与假手术组比较,模型组大鼠神经缺损评分明显升高(P<0.01),脑组织病理损伤严重,血清NSE水平明显升高(P<0.01),脑组织中IL-10,TGF-β_1水平明显降低(P<0.01);与模型组比较,凌霄花总黄酮高剂量组可明显改善模型大鼠的神经缺损症状,降低神经缺损评分(P<0.05);凌霄花总黄酮高、低剂量组和阿司匹林组能明显减轻脑组织病理损伤程度(P<0.01,P<0.05),降低血清中NSE水平(P<0.05),升高脑组织中IL-10,TGF-β_1水平(P<0.01,P<0.05)。结论:凌霄花总黄酮对短暂性脑缺血再灌注大鼠有一定的保护作用,可能与降低NSE水平,升高抗炎细胞因子IL-10,TGF-β_1的水平,减轻炎症损伤有关。Objective: To observe the protective effects of total flavonoids from Campsis Flos (CFTF) on cerebral ischemia reperfusion injury in rats. Method: Sixty SPF grade Wistar rats were randomly divided into sham operation group, model group, positive drug group (aspirin, 50 mg·kg^-1), CFTF high dose and low dose groups (200, 100 mg·kg^-1). Models of transient middle cerebral artery occlusion (tMCAO) were established in all the rats except those in sham operation group. The behavioral injury was detected by neurologic deficit scores (NDS); the cerebral histopathology was detected by HE staining; and the levels of neuron specific enolase (NSE) in serum, interleukin(IL)-10, and TGF-β1 in brain homogenate were detected by ELISA, to discuss the protective effect of CFTF on cerebral ischemia reperfusion injury. Result: As compared with sham operation group, the neurologic deficit scores were significantly higher in rats of model group (P〈0.01), with serious cerebral histopathology injuries; and the level of NSE in serum was significantly increased (P〈0.01); the levels of IL-10, and TGF-β1 in brain homogenate were decreased dramatically (P〈0.01). As compared with the model group,the neurologic deficit symptoms were significantly improved in CFTF high dose group (200 mg·kg^-1), with decreased neurologic deficit scores (P〈0.05); cerebral histopathology injury was significantly decreased(P〈0.01, P〈0.05), NSE level in serum was decreased (P〈0.05), and levels of IL-10, TGF-β1 in brain tissues were increased (P〈0.01, P〈0.05) in CFTF high dose and low dose groups (200, 100 mg·kg^-1) and aspirin group. Conclusion: CFTF had the protective effects on tMCAO rats by decreasing the level of NSE, increasing the levels of anti-inflammation cytokines (IL-10 and TGF-β1), and attenuating the inflammation injury.
