摘 要:目的观察补肾益冲抗衰汤对卵巢储备功能下降模型大鼠血清内分泌及抗苗勒管激素(AMH)、抑制素B(INHB)的影响。方法选取40只8周龄雌性SD大鼠,分为正常对照组、模型组、补佳乐组、补肾益冲抗衰汤组,各10只,除正常对照组外,其余各组采用雷公藤多苷片+相应药物制作卵巢储备功能下降大鼠模型。通过阴道涂片观察各组大鼠动情周期,光镜下观察卵巢组织形态学变化,酶联免疫法检测血雌二醇(E2)、卵泡刺激素(FSH)、黄体生成素(LH)、AMH、INHB水平。结果补肾益冲抗衰汤能维持大鼠动情周期4~5天,促进卵泡发育成熟、改善内分泌水平,与模型组比较,E2上升[(21.51±1.26)pg/m L比(9.81±1.18)pg/m L,P〈0.05],FSH下降[(6.57±1.88)m IU/m L比(9.59±2.17)m IU/m L,P〈0.05],FSH/LH降低[(2.27±0.44)比(3.36±0.82),P〈0.05];升高AMH[(114.45±11.11)ng/L比(100.27±9.17)ng/L,P〈0.05]和INHB[(56.91±1.74)ng/L比(26.28±6.19)ng/L,P〈0.05]水平。结论补肾益冲抗衰汤能够改善卵巢储备功能下降(DOR)大鼠性激素水平,升高AMH、INHB水平,并减少闭锁卵泡,改善大鼠卵巢储备功能。Objective To observe the influence of Bushen Yichong Kangshuai Decoction(BSYCKSD) on endocrine and anti-Mullerian(AMH) and inhibin B(INHB) expressions of diminished ovarian reserve model rats.Methods Eight-week old 40 healthy female rats as the research object were divided into normal control group,model group, prognova group, BSYCKSD group, with 10 rats in each group.Except for normal control group,triptery-gium wilfordii and corresponding medicinewere used to set up diminished ovarian reserve model in rats.The estrous cycle was determined through the observation of vaginal smear. The morphological changes of ovarian tissue was observed by light microscopic. ELISA method was for measuring serum estradiol(E2), follicle stimulating hormone(FSH), luteinizing hormone(LH), AMH and INHB. Results In BSYCKSD group, the estrous cycle of rats lasted 4to 5d, the development of mature follicles was promoted, and the endocrine levelswas improved compared with model group(E2: 21.51 ±1.26pg/m L vs 9.81 ±1.18pg/m L; FSH: 6.57 ±1.88 m IU/m L vs 9.59 ±2.17 m IU/m L; FSH/LH:2.27±0.44 vs 3.36±0.82; all P〈0.05) and the levels of AMH(114.45±11.11 ng/L vs 100.27±9.17ng/L,P〈0.05) and INHB(56.91 ±1.74ng/L vs 26.28 ±6.19ng/L,P〈0.05) were increased. Conclusion BSYCKSD can improve sex hormone levels in rats with diminished ovarian reserve, increase the level of AMH and INHB, and reduce the follicu-lar atresia, resulting in enhancing the reserve function of ovary.
