温阳活血法对寒凝血瘀证荷CT-26小鼠肺组织MMPs及ERK的影响

摘　　要：目的观察寒凝血瘀状态及温阳活血法对肿瘤转移靶组织微环境的影响。方法建立寒凝血瘀证荷瘤小鼠复合动物模型。将48只Balb/c小鼠随机分为空白对照组、单纯荷瘤组、寒凝血瘀荷瘤组、温阳活血治疗组,每组12只,分别造模,Western blot方法检测各组小鼠肺组织MMPs及ERK的表达。结果与单纯荷瘤组比较,寒凝血瘀荷瘤组小鼠ERK1/2、MMP-2表达均升高[ERK1/2 RI值:(1.40±0.01)比(0.98±0.03),P<0.05];[MMP-2 RI值:(1.26±0.05)比(1.00±0.04),P<0.05];与寒凝血瘀荷瘤组比较,温阳活血治疗组小鼠ERK1/2、MMP-2表达均降低[ERK1/2 RI值:(1.18±0.01)比(1.40±0.01),P<0.05];[MMP-2 RI值:(1.01±0.05)比(1.26±0.05),P<0.01]。与寒凝血瘀荷瘤组比较,温阳活血治疗组小鼠MMP-12表达升高[MMP-12 RI值:(1.23±0.03)比(1.08±0.06)(P<0.05)]。结论温阳活血法通过下调荷瘤小鼠肿瘤转移靶器官肺组织ERK1/2、MMP-2表达及上调MMP-12从而影响肿瘤基质及肿瘤血管生成,进而减少转移灶形成的发生率。Objective To investigate the effect of Chinese medicine cold coagulation and blood stasis syndrome and Yang-warming and blood-activating therapy on tumor metastasis. Methods Forty-eight Balb/c mice were randomly divided into 6 groups： blank control group, tumor-bearing group, tumor-bearing＋cold coagulation and blood stasis syndrome group（control group with tumor bearing）, tumor-bearing ＋cold coagulation and blood stasis syndrome＋herb medicine group（treatment group）, with 12 mice in each group. Except for blank control group, cold coagulation and blood stasis syndrome model was established in other groups. The expression of MMPs and ERK in lung tissue was determined by western blot assay. Results Compared with tumor-bearinggroup, ERK1/2 and MMP-2 of control group with tumorbearing were higher（ERK1/2 RI value：1.40 ±0.01 vs 0.98 ±0.03; MMP-2 RI value：1.26±0.05 vs1.00±0.04; all P〈0.05）. Compared with control group with tumorbearing, treatment group had decreased ERK1/2（RI value： 1.18±0.01 vs 1.40±0.01, P〈0.01） and MMP-2（RI value： 1.01±0.05 vs 1.26±0.05; all P〈0.01）,but increased MMP-12 level（RI value： 1.23±0.03 vs 1.08±0.06,P〈0.05）. Conclusion Yang-warming and bloodactivating therapy can decrease the expression of ERK1/2 and MMP-2 and increase MMP-12 of lung tissuein tumor-bearing mice to influence tumor stroma and the formation of tumor vessels and consequently reduce the incidence of tumor metastasis.
